ObjectiveA study on the neuroprotective effects of lead-removing Fuzheng recipe on lead poisoning rats was conducted to investigate the neuroprotective effects of lead-removing Fuzheng recipe on lead poisoning rats through Keap-1-Nrf2-ARE pathway involved in oxidative stress regulation.MethodsThe rat model of lead poisoning was established by drinking lead acetate water.The 53clean-grade SD rats were randomly divided into blank groups?n=8?after one week of adaptive feeding,drinking distilled water;the lead poisoning model group?n=45?,drinking 0.2%lead acetate water.After 40 days of exposure,five blood samples were randomly selected from the lead poisoning model group to detect blood lead content to confirm the success of the modeling.After successful modeling,the model group of lead poisoning was randomly divided into five groups:model group,western medicine group,high dose group of traditional Chinese medicine,middle dose group of traditional Chinese medicine and low dose group of traditional Chinese medicine.The rats in the six groups were given distilled water normally,among which the western medicine group was given injection of calcium ediate?CaNa2EDTA??50.7 mg/kg d?,7courses of treatment?3 consecutive injections days,4 stops?,and 4 courses of treatment;the traditional Chinese medicine treatment group was given 10 ml/kg.d of different concentrations?1.75,0.875,0.4375 g/ml?of lead-removing Fang extract.During this process,the blank group and the model group were given 0.9%saline 10 ml/kg.d gavage.The body weight changes of rats were detected after 4 weekends of treatment,and the spatial learning and memory ability of rats in each group was detected by Morris water maze experiment.The contents of blood lead,liver lead and kidney lead were detected by atomic absorption spectrophotometer;the content of superoxide dismutase?SOD?,malondialdehyde?MDA?,glutathione enzyme?GSH?were detected by xanthine oxidase method,TBA method and colorimetric method;and the protein expression of brain tissue was detected by immunohistochemistry and Western blotting method.Results1.Weight changes in rats before and after the experiment:compared with the blank group,the weight gain in the model group decreased significantly?p<0.01?,while the weight gain in the western medicine group and the high,medium and low dose groups of traditional Chinese medicine increased significantly compared with the model group?p<0.01?.2.Levels of lead in rats in each group were significantly higher than those in the blank group?p<0.01?.Compared with the model group,the levels of lead in liver tissue,liver tissue and kidney tissue were significantly lower in western medicine group and Chinese medicine group?p<0.01?.3.Results of behavioral testing in rats:compared with the blank group,the duration of escape latency in the model group increased significantly and the number of times crossing the safe platform decreased significantly?p<0.01?.Compared with the model group,the duration of escape latency in the western medicine group and the high,middle and low dose groups of traditional Chinese medicine decreased significantly and the number of times crossing the platform increased significantly?p<0.01?.4.Results of biochemical indexes of brain tissue in 4 rats:compared with blank group,the SOD?GSH content in brain tissue of model group was significantly decreased and the MDA content was significantly increased?p<0.01?.Compared with model group,the SOD?GSH content in brain tissue of western medicine group and Chinese medicine group was significantly increased and the MDA content was significantly decreased?p<0.01?.High levels of SOD?GSH in brain tissue of low-dose Chinese medicine,decreased MDA content?p<0.05?.5.Rats brain tissue Keap-1-Nrf2-ARE pathway related protein expression:compared with the blank group,the Keap-1 protein expression of the model group increased significantly,and the HO-1?Nrf2 protein expression decreased significantly?p<0.01?.Conclusion1.Lead-driven square can effectively reduce lead content in blood,liver and kidney of rats with lead poisoning.2.Lead-driven square can effectively improve the learning and memory ability of lead poisoning rats.3.Lead-driven square plays a neuroprotective role by inhibiting oxidative stress response in lead poisoning rats,regulating Keap-1-Nrf2-ARE pathway related protein expression.
