Objective To study the therapeutic effect of pirfenidone(PFD)on renal interstitial fibrosis in rats and its influnce on the expression of Dab2.Methods Forty-eight standard SPF male SD rats were randomly divided into 3 groups:control group,model group and pirfenidone treatment group.Left ureteral ligation was performed in model group and pirfenidone treatment group to replicate the renal interstitial fibrosis model,and the model of unilateral ureteral obstruction(UUO)was made.The rats in the treatment group were given pirfenidone 250 mg/(kg·d)by gavage,and the rats in the control group and model group were given 1% sodium hydromethylcellulose solution by gavage.Eight rats in each group were sacrificed on day 7th and 14 th after the operation,and the obstructed kidney was taken.The pathological changes of renal tissues in each group were observed by HE and Masson staining,and the expression levels of Dab2 m RNA were detected by real-time quantitative polymerase chain reaction(qrt-pcr).Results HE staining results showed that the control group did not show renal fibrosis,model group showed rat renal interstitial fibrosis and inflammatory cell infiltration.The degree of renal fibrosis and inflammatory cell infiltration in treatment group was significantly lower than that in model group.Masson staining results showed that The renal tissue structure of the control group was normal,and there was no obvious collagen fiber in the interstitial.After the model group was established,a large number of collagen fibers were deposited.At the 14 th day,diffuse blue stained collagen fibers were basically formed in the model group.Compared with the model group,the blue stained collagen fibers in the renal interstitial of the pirfenidone treatment group were significantly reduced.In model group,the index of renal interstitial damage and the score of collagen relative area were higher than those in control group(P < 0.05),and there were significant changes after treatment with pirfenidone(P < 0.05).The relative expression of DAB2 m RNA in model group was higher than that in control group(P <0.05);the relative expression of DAB2 m RNA in treatment group was higher than that in model group(P < 0.05);the relative expression of DAB2 m RNA in control group was also higher than that in control group(P < 0.01);the difference was statistically significant.Conclusions Pirfenidone can significantly improve the degree of renal fibrosis in UUO rats,and its mechanism may be related to the relative expression of Dab2 in renal tissue.
