Objective: The tissue and organ damage and dysfunction of patients due to diseases,trauma and other reasons,the development of tissue engineering provides a new method for wound restoration.The experiment started with the differentiation characteristics of adipose-derived stem cells(ADSCs),formed them into cell sheets,combined with nano-hydroxyapatite / collagen scaffolds,and inoculated with adipose stem cells.The suspensions were compared for the ability of adhesion and osteogenic differentiation in vitro.Methods: 1.Take adipocytes from liposuction in healthy people,extract ADSCs,and conduct in vitro cultivation and identification.2.Preparation of nHAC scaffold.3.Construct ADSCs cell sheets and observe them by histological staining to induce osteogenesis.4.Set up the ADSCs suspension-nHAC scaffold group,and the ADSCs cell sheets-nHAC scaffold group to determine the early cell adhesion rate and the expression of osteogenesis,and observe the structure under scanning election microscope.Results: 1.The cell growth curve was nearly "S" type.The flow phenotype identification of CD44 and CD105 were positively expressed,and CD45 were negatively expressed.The results of osteogenesis and adipogenesis confirmed that the extracted cells were ADSCs.2.Hematoxylin-eosin staining of the cell sheets showed a stratified cell arrangement,and large slices of ALP staining were seen in osteogenesis induction.3.In the experiments,compared with other groups,the ADSCs cell sheets with nHAC scaffold group showed good cell adhesion rate,and the expression of ALP significantly increased,with statistical significance.Conclusion: ADSCs were induced into the cell sheets,and co-cultured with nano-hydroxyapatite/collagen scaffolds,cell sheets showed better cell adhesion rate and the higher expression of osteogenesis.
