Focusing on building varies kinds of tissue-engineered bone,bone tissue regeneration aims at repairing bone structure and function.Bone tissue regeneration is one of the main methods to treat the increasing orthopedic diseases all over world.To smoothly process this work,the selecting and synthesizing of materials,designing and building of scaffolds and in vivo transplantation of biomedical engineering are all involved in.Mesoporous bioactive glass(MBG)has performed excellent osteoinductivity and osteoconductivity in bone tissue regeneration for its regular structure and rather big specific surface area and pore volume.Because of the outstanding biocompatibility and bioactivity,Collagen and Chitosan are the most frequently-used in tissue regeneration among natural biomaterials.To research whether it is feasible to take MBG loaded with testosterone(MBG/T)as particle for bone regeneration,we first synthesized MBG by the method of sol-gel;then,we detected the control release of MBG/T by studying the adsorption and release of testosterone from MBG.Thirdly,the influence of MBG/T on the proliferation,differentiation and mineralization of MC3T3-E1 cells were studied by in vitro cultivation.The results presented that MBG/T did do evident promotion on cell proliferation,differentiation and mineralization,which were complete consistent with the obvious expression of marker genes and proteins related with bone formation.Based on these works,we did further study by freeze-drying Collagen and Chitosan to build three-dimensional scaffold(Collagen-Chitosan),then the Collagen-Chitosan scaffold was used to load MBG to make final complex scaffold(Collagen-Chitosan-MBGs).After that,the influence of Collagen-Chitosan-MBGs on the proliferation,differentiation and mineralization of bMSC cells were studied by in vitro cultivation.The results showed that Collagen-Chitosan-MBGs scaffolds did do evident promotion on cell proliferation,differentiation and mineralization,which were complete consistent with the obvious expression of marker genes and proteins related with bone formation.All in all,the conclusions drew from our paper show great potential in the study of bone tissue regeneration based on Collagen,Chitosan and MBG.
