Objective: In this study,a glycyrrhizin-conjugated bovine serum albumin-containing active ingredient carrier for luteolin was constructed,and luteolin-loaded nanoparticles(GL-BSA-Lut-Nps)were prepared and selected for liver cancer Bel-7402 cells nanoparticles with the best inhibitory activity and explore their antitumor activity in vitro.Methods: 1)Chinese medicine active carrier GL-BSA was prepared by chemical coupling method and lyophilized after dialysis;2)Desolvation method was used to prepare 5 kinds of drug-loaded nanoparticles under prescription and freeze-drying;3)The MTT method is used to detect the activity of 5 kinds of GL-BSA-Lut-Nps anti-liver cancer Bel-7402 cells and to select the best prescription for inhibitory activity;4)Flow cytometry to detect cell cycle arrest and apoptosis;5)Measure the solubility of GL-BSA-Lut-Nps and luteolin with an ultraviolet spectrophotometer;6)Detection of GL-BSA-Lut-Nps targeting Bel-7402 cells with fluorescently labeled drugs;7)The metabolism of Bel-7402 cells by GL-BSA-Lut-Nps was studied by 1 H NMR.Results: 1)Successfully prepared 5 kinds of GL-BSA-Lut-Nps lyophilized powder;2)The IC50 values of five parallel samples of GL-BSA-Lut-Nps treated liver cancer Bel-7402 cells for 24 hours were 10.730 ± 1.120,6.079 ± 0.580,3.602 ± 1.220,1.999 ± 0.880 and 3.443 ± 0.990 mg/mL;3)GL-BSA-Lut-Nps-4 showed S-phase arrest of Bel-7402 cells,and the apoptosis induction rate was 65.70%;4)The solubility of GL-BSA-Lut-Nps-4 was 1.5 × 10-2 g/mL,and the solubility of Lut was 1.7 × 10-5 g/mL;5)Fluorescence assay results showed that the experimentally constructed GL-BSA-Lut-Nps-4 has better tropism towards the Bel-7402 cells;6)GL-BSA-Lut-Nps-4 had obvious metabolic regulation effect on Bel-7402 cells,mainly involving aerobic oxidation,phospholipid metabolism,energy metabolism,amino acid metabolism and other metabolic processes.Conclusion: In this paper,desolvation method was used to successfully prepare glycyrrhizic acid-conjugated bovine serum albumin-loaded luteolin nanoparticles,and GL-BSA-Lut-Nps-4 nanoparticles prepared under the best prescription were selected.Liver cancer Bel-7402 cells have increased activity and water solubility,providing a certain theoretical basis for Lut formulation and development of new and effective Chinese medicine preparations for the treatment of liver cancer.
