Objectives To explore the effect of mirna-155-3p on apoptosis and cell cycle of drug-resistant colon cancer cells.Methods Four human colorectal cancer cell lines SW480,HCT-15,HT29 and HCT116 were established by the combination of high dose intermittent induction.The SW480,HCT-15,HT29 and HCT116 and their corresponding oxaliplatin resistant cell lines were detected by Q-PCR,and the most different cell lines were selected.SW480 cells were randomly divided into blank group,experimental group and NC group.The blank group was added with PBS,the experimental group with miRNA-155-3p inhibitor,the NC group with miRNA NC,the transfection efficiency was detected by Q-PCR,the expression of BCL2 and Caspase-3 protein in cell line was detected by wertern blot,and the cell cycle of each group was detected by flow cytometer.Results The RI index of moderately resistant colon cancer cells was 10.64,9.63,6.93,6.33,respectively,The relative expression of miRNA-155-3p in SW480/OXA,HCT116/OXA,HT29/OXA,HCT15/OXA were 10.049±1.583,2.172±0.334,2.791±0.250,2.661±0.409,which was higher than that in the four cancer cell lines(P<0.05),indicating that the increased expression of miRNA-155-3p might be related to the drug resistance of colon cancer cells;miRNA-155-3p in the experimental group of transfection inhibition plasmid,the difference was statistically significant(P<0.05),indicating that the transfection was successful;The relative expression of BCL2 protein in the experimental group was 0.639±0.006,in the blank group and NC group was 0.856±0.008,0.880±0.017,the difference was statistically significant(P<0.05);the relative expression of Caspase-3 protein in the experimental group was 1.033±0.041,in the blank group and NC group was 0.827±0.018,0.857±0.009,the difference was statistically significant(P<0.05).Inhibition of miRNA-155-3p can inhibit BCL2 and promote Caspase-3 expression,thus increasing the apoptosis of drug-resistant colon cancer cells;The proportion of G0/G1 phase cells in the experimental group was 57.260%,higher than 43.06% in the blank group and 42.74% in the NC group,the difference was statistically significant(P<0.05),indicating that inhibition of miRNA-155-3p can effectively interfere with the cell cycle of drug-resistant colon cancer cells.Conclusions Inhibition of miRNA-155-3p can effectively interfere with the cell cycle of drug-resistant colon cancer cells and promote apoptosis.Inhibition of miRNA-155-3p may be an effective way to solve the drug resistance of tumor cells.Figure 3;Table 6;Reference 183...
