intestinal environment of type 2 diabetic mice,and the regulation effect of Trichosanthes pericarpimm on the intestinal flora of type 2diabetic mice..The specific research content and results are as follows:1、The hypoglycemic effect of Trichosanthes pericarpimm on type 2 diabetic mice and the effect of changes in intestinal flora on blood glucose in diabetic mice were studied.Injecting streptozotocin(STZ)to induce a mouse model of type 2 diabetes and administering high-medium-low-dose melon peel suspensions;at the same time,the intestinal microflora was isolated from the feces of the high-dose Trichosanthes pericarpimm gavage group mice,and the fecal bacteria extract was prepared and administered to the diabetic mice.The hypoglycemic activity of Trichosanthes pericarpimm was studied by analyzing the body weight,fasting blood glucose,blood lipid levels,serum insulin,insulin resistance index,insulin sensitivity index and organ index of mice.The results showed that Trichosanthes pericarpimm and faecal extracts were administered by gavage to inhibit fast weight gain and organ enlargement in diabetic mice,reduce fasting blood glucose levels in diabetic mice,and improve blood lipid levels in diabetic mice.In addition,Trichosanthes pericarpimm and faecal extracts can significantly weaken the insulin resistance of diabetic mice,and reduce the serum insulin level,insulin resistance index and insulin sensitivity index of diabetic mice.The analysis results show that Trichosanthes pericarpimm may improve the glucose and lipid metabolism of diabetic mice by changing the intestinal microbiota of diabetic mice,and achieve hypoglycemic and insulin resistance in type 2 diabetic mice.2、The effects of Trichosanthes pericarpimm and faecal extract on the intestinal microecology of diabetic mice and the molecular mechanism of hypoglycemic effect were studied.The fecal p H of the diabetic mice before and after gavage were compared,and the ammonia mass fraction of the feces of the mice,the serum lipopolysaccharide-binding protein level of the mice,and the liver adenosine momophosphate activated protein kinase(AMPK)and Sirtuin 1(SIRT1)levels were analyzed.The number of Bifidobacterium,Bacteroides,Lactobacillus and Clostridium coccoides in the intestinal tract of diabetic mice before and after gavage were compared by plate counting method,and the mechanism of hypoglycemic effect of Trichosanthespericarpimm and fecal bacteria extract was analyzed by RT-PCR.The results showed that Trichosanthes pericarpimm and faecal extracts were administered to the stomach to improve the intestinal environment of diabetic mice,and to reduce the stool p H and ammonia content of the stool.Trichosanthes pericarpimm and faecal extracts alao increase the number of beneficial bacteria Bifidobacterium,Bacteroides,and Lactobacillus in the intestine of diabetic mice,and reduce the number of harmful bacteria Clostridium coccoides.In addition,Trichosanthes pericarpimm and faecal extracts reduced the serum lipopolysaccharide binding protein content of diabetic mice and increased the levels of liver AMPK and SIRT1 in diabetic mice.The results of RT-PCR analysis showed that the Trichosanthes pericarpimm and faecal extract could regulate the expression of glucose transporter 1(GLUT1),glucose transporter 4(GLUT4)and phosphatidylinositol 3-kinase(PI3K)genes in the liver of diabetic mice,and at the same time inhibit the expression of sodium-glucose cotransporter 1(SGLT1),forkhead transcription factors of the O class 1(FOXO1),phosphoenolpyruvate carboxykinase(PEPCK)and Glucose-6-phosphatase(G6Pase)genes,thereby regulating glucose metabolism and improving insulin resistance in diabetic mice.3、The effects of Trichosanthes pericarpimm and faecal extract on the intestinal microflora of type 2 diabetic mice were studied.Based on the 16 Sr DNA high-throughput sequencing method,the effect of Trichosanthes pericarpimm and faecal extract gavage on the intestinal microbial diversity of diabetic mice was analyzed.The differences in intestinal flora of each group of mice were also compared.OTU cluster analysis,principal component analysis,intestinal bacterial community composition analysis,and species abundance cluster analysis were performed on the intestinal flora of each group of mice.The results showed that the number of OTUs in the diabetic mice after gavage was significantly higher than that in the diabetic model control mice,and the diversity was also significantly increased;Principal component analysis showed that the composition of diabetic mice after gavage was more similar to that of normal mice;The community composition analysis showed that the intestinal flora structure of diabetic mice after gavage was significantly changed,and the cluster analysis of species abundance showed that the intestinal flora structure of diabetic mice after gavage was closer to that of normal mice.The analysis results revealed that the Trichosanthes pericarpimm and faecal extracts both regulated the intestinal microbiota of type 2diabetic mice,confirming that the hypoglycemic effect of Trichosanthes pericarpimm on type 2 diabetic mice was related to its effect on changing the intestinal microbial population. |
