Objective: Objective to investigate the anti-atherosclerotic effect of Allisartan and its effect on Interleukin-6(IL-6)and Lipoprotein-associated phospholipase A2(Lp-PLA2).Methods:1.Fifteen healthy purebred New Zealand male white rabbits were randomly divided into the following three groups after one week of adaptive feeding: the normal control group was fed with ordinary feed;the high-fat diet group was fed with high-fat feed(feed formula: 1% cholesterol + 6% Peanut oil + 93% basal feed);Allisartan intervention group were fed with high-fat diet and Allisartan 40 mg /(kg · d).2.Thirteen weeks later,blood was collected and the rabbits were sacrificed.The thoracic aorta was retained,and pathological changes were observed by HE staining and Masson staining.Enzyme method for total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),and enzyme-linked immunosorbent assay method to determine IL-6,Lp-PLA2 level.3.Statistical analysis was performed by SPSS22.0 statistical software.The data were expressed by x±s,and comparison between groups was performed by one-way analysis of variance.P <0.05 was statistically significant.Results: 1.During the experiment,there was no statistical difference in the food intake of the three groups of rabbits(P> 0.05).The body weight,TC,TG,and LDL-C of the high-fat diet group and the Allisartan intervention group were higher than those of the normal control group.HDL-C levels were higher than those of the normal control group.The difference was statistically significant(P<0.05).Compared with the high-fat diet group,there was no significant difference in the levels of TC,TG,LDL-C,and HDL-C in the Allisartan intervention group(P> 0.05).2.The HE staining results showed that Allisartan can reduce the atherosclerotic(AS)progression of rabbit thoracic aorta,foam cells in the high-fat diet group in the Alissartan intervention group were significantly reduced,and intimal thickening was lighter.The intimal/media thickness ratio of the Alissartan intervention group was higher and the fat diet group was significantly reduced(P<0.05).Masson staining showed that vascular fibrosis was significantly reduced in the high-fat diet group in the Allisartan intervention group,and blue-stained collagen fibers were reduced.3.Enzyme-linked immunosorbent assay results showed that the serum IL-6 and Lp-PLA2 expression levels in the high-fat diet group and the Alissartan intervention group were higher than those in the normal control group(P <0.05).Compared with IL-6,Lp-PLA2 expression levels were reduced(P <0.05).Conclusions:1.Allisartan does not affect the TC,TG,LDL-C,HDL-C levels of AS rabbits.2.Allisartan can reduce IL-6 and Lp-PLA2 levels,may have anti-inflammatory effects.3.Allisartan can reduce the atherosclerosis progression in the thoracic aorta of rabbits,and its mechanism may be related to down-regulating the inflammatory response.
