Objective: To investigate the difference in expression levels of the cholecystokinin receptor(CCKR)pathway in cholangiocarcinoma and adjacent tissues and its role in the pathogenesis of cholangiocarcinoma.Methods: Twenty-four patients with bile duct cancer diagnosed in the affiliated hospital of their school from August 2015 to June 2019 were collected.KEGG analysis and GO function analysis were performed based on ADVID and PATER platforms after detecting the gene expression of tumor tissues of 6 patients with biochip,and the construction of corresponding PPI network was completed.The expression levels of ITGB1,CCND1,SERPINE1,and CCKAR in the cancer and adjacent tissues of patients in the CCKR pathway were determined by real-time fluorescent quantitative nucleic acid amplification;the downstream products of CCKR in the serum of patients were detected by enzyme-linked immunosorbent assay(ELISA)Compared with normal people and patients after surgery.Results: There were significant differences in gene expression among individuals with cholangiocarcinoma.Differential genes are mostly enriched in signaling pathways such as CCKR,PI3K-Akt,cell bridge regulation,and cell cycle changes.At the tissue level,the expression levels of CCND and CCKAR on the CCKR pathway were significantly increased in patients(P <0.05),and the expression of ITGB1 and SERPINE showed an upward trend(P> 0.05).The CCKAR content in patients' serum was significantly higher than normal(P < 0.05).Conclusion: The expression of CCKR signaling pathway in tumor tissues and serum of patients with cholangiocarcinoma is significantly up-regulated.Considering its possible pathogenic mechanism,it acts on important sites such as cell bridges,nucleic acid binding proteins and transcription factors,and then changes the life cycle of tumor cells.Effect,affecting the prognosis of patients.
