Objective:Polycystic ovary syndrome(PCOS)is the most common reproductive endocrine and metabolic disease among women of childbearing age.Micro RNA is involved in many biological processes,including inflammation,fat formation and insulin signaling,etc.Important biological functions.This article intends to evaluate the effects of miRNA-21-5p and miRNA-27a-3p in the blood on PCOS-related indicators,especially metabolic indicators,to explore the correlation with PCOSMethods:1.142 PCOS patients who were first diagnosed in the Department of Reproductive Medicine of the Second Hospital of Hebei Medical University from July 2019 to December 2019 were selected.According to WHO's diagnostic criteria for obesity,it is divided into three groups according to body mass index(BMI);normal body reorganization: 31 cases;super reorganization: 61 cases;obesity group: 50 cases.According to the level of testosterone(T),it is divided into hyperandrogenism(HA)and non-hyperandrogenism(NHA).SPSS 22.0 software was used to perform statistical analysis on clinical indications and biochemical indicators,and to investigate whether there were correlations between the indicators.The difference was statistically significant at P <0.05.2.8 people in the normal weight PCOS group and 15 people in the obese PCOS group(obese people both weight loss> 5%),through the design of primers,the expression levels of miRNA-21-5p and miRNA-27a-3p in the blood were measured by real-time fluorescence quantitative method.Compare the difference in expression of miRNA between obesity and normal body reorganization,before and after weight loss,and whether it is correlated with metabolic indicators.Results:1.(1)General data: The body weight,BMI,body fat rate,visceral fat level,waist circumference,waist-to-hip ratio of the obesity group were higher than that of overweight group,which was higher than that of normal weight group(P <0.05).(2)Comparison of liver function indexes: The differences among the three groups of ALT and AST were statistically significant,and the levels of ALT and AST in obesity and overweight group were higher than normal weight group(P <0.05);the difference between DBIL,IBIL and ALP among the three groups was not statistically significant(P > 0.05).(3)Comparison of lipid metabolism indicators: The TG?HDL?LDL?ApoA?ApoB levels among the three groups were statistically significant(P <0.05);The levels of TC?TG?LDL?ApoB in obesity group were higher than normal weight group,the levels of TG and ApoB in overweight group were higher than the normal weight group,and the difference was statistically significant(P <0.05).The levels of HDL in normal weight group was higher than the overweight and obesity group,and the difference between the two groups was statistically significant(P <0.05).The level of ApoA in normal weight group is higher than overweight group,the difference is statistically significant(P <0.05).(4)Comparison of glycometabolism indexes: The differences in HbA1c?FINS and IR between the three groups were statistically significant(P <0.05).The HbA1 c and FINS in the overweight and obesity group were significantly higher than those in the normal weight group;the levels of FBG in the obesity group was significantly higher than that in the normal weight and overweight group,the difference was statistically significant(P <0.05).IR in the obesity group was higher than that of overweight group and higher than that of normal weight group,and the comparison between two groups was statistically significant(P <0.05).(5)Comparison of reproductive hormones: The LH levels among the three groups were statistically significant(P <0.05).LH was higher in the normal weight group than in the overweight group,and the difference was statistically significant(P<0.05).The AMH level of normal weight gruop was higher than that of overweight group than that of obesity group,the difference between normal weight group and obesity group was statistically significant(P <0.05).2.The TC,TG,LDL and ApoB in the HA group were higher than those in the NHA group,while the HDL and ApoA were lower than the NHA group.3.BMI is positively correlated with ALT,AST,TC,LDL,ApoB,HbA1 c,FINS,IR,but negatively correlated with HDL,ApoA,AMH,FINS is positively correlated with ALT,AST,TC,LDL,ApoB,HbA1 c,IR,and HDL and ApoA were negatively correlated,and T was positively correlated with LDL,Apo-B and FINS.The difference was statistically significant(P <0.05).4.The expression levels of miRNA-27a-3p and miRNA21-5p in normal body were higher than those in the obese group.After weight control,the expression levels of miRNA21-5p increased compared with the previous ones.5.miRNA-27a-3p and miRNA-21-5p were negatively correlated with BMI,body fat percentage,visceral fat level,LDL,ApoB,FINS,IR,and positively correlated with HDL.Conclusions:1.PCOS patients interact with each other.Obese PCOS patients are more likely to have abnormal liver function,hyperlipidemia,hyperinsulinemia,IR and other metabolic abnormalities than normal-weight PCOS patients.In PCOS patients,HA is more prone to abnormal lipid metabolism than NHA.2.miRNA-21-5p and miRNA-27a-3p may be involved in the regulation process of obesity in PCOS patients,and control their weight to increase their expression levels;and it was speculated that the expression level of PCOS was correlated with glucose and lipid metabolism.
