Objectives The relationship between Cdc2 as an upstream regulatory gene of BAD and the occurrence and development of cervical cancer was confirmed through experiments,and the expression and clinical significance of Cdc2,BAD,and Emi2 in different cervical lesions and their correlation with high-risk HPV infection were studied This will provide new molecular targets and experimental directions for the research,diagnosis and treatment of cervical cancer.Methods A total of 102 surgical specimens from Tangshan Workers' Hospital from September 2017 to January 2019 were selected as the research object,Divided into four groups.DNA typing of high-risk HPV(HR-HPV)in patients with HPV detection,including HPV16,18,31,33,35,39,45,51,52,56,58,59,68,73 types,Summarized into three categories are HPV16,HPV18 and 12 other high-risk subtypes of HPV.The expression of Cdc2 protein and BAD protein in each group was detected by immunohistochemistry.Real-time fluorescence quantitative PCR technology was used to detect the expression level of Emi2 gene in each group.And analyze the correlation between the clinicopathological characteristics of cervical cancer tissues and the results of Cdc2 protein,BAD protein and Emi2 gene in cervical cancer.The correlation between Cdc2 protein and BAD protein and the correlation between Cdc2 protein,BAD protein and Emi2 gene and HR-HPV were analyzed.The difference between the expression levels of Cdc2 protein,BAD protein and Emi2 gene expression in each group was analyzed by ANOVA;the measurement data was expressed as mean ± standard deviation((?)x±s);Cdc2 protein,BAD protein and Emi2 gene were obtained by detection The relationship between the results and the clinicopathological characteristics of cervical cancer tissues was determined by chi-square test;the comparison of Emi2 gene between the two groups was by independent sample t test.The correlation between Cdc2 protein,BAD protein,Emi2 gene and HR-HPV was analyzed by Spearman correlation.Results The positive rate of Cdc2 protein detected by immunohistochemistry in the normal cervical tissue group,CINI group,CIN?-? group,and cervical cancer group was 7.6%,26.1%,48.0%,and 82.1%,respectively.Academic significance(?2=53.680,P<0.001).The positive rate of BAD protein detected by immunohistochemical method in the normal cervical tissue group,CINI group,CIN?-? group,and cervical cancer group was 76.9%,69.6%,52.0%,and 28.6%,respectively.Academic significance(?2=31.212,P=0.012).The positive rate of Emi2 gene detected by real-time fluorescent quantitative PCR technology in normal cervical tissue group,CINI group,CIN?-? group and cervical cancer group was 11.5%,21.7%,28.0%,82.1% respectively.Obvious statistical significance(?2=51.667,P<0.001).In the relationship between the clinical and pathological characteristics of patients with cervical cancer: Cdc2 in the lymph node metastasis group(?2=6.97,P=0.018),pathologically graded poorly differentiated tissue group(?2=12.14,P=0.001),clinical stage III The expression rate was higher in the group(?2=8.43,P=0.005),but there was no significant difference in expression rate between different ages(?2=3.31,P=0.078).The expression rate of BAD in the pathologically graded poorly differentiated tissue group(?2=18.31,P=0.001),clinical stage III groups(?2=9.14,P=0.013)was higher,and the group with lymph node metastasis(?2=4.97,P=0.073),no significant difference in expression rate between different ages(?2=2.56,P=0.112).Emi2 expression was higher in histologically poorly differentiated tissues(t=8.87,P=0.018)and lymph node metastasis(t=6.55,P=0.021),but in different age groups(t=0.48,P=0.622)There was no significant difference in expression between clinical staging groups(t=0.62,P=0.431).The HR-HPV infection rate was significantly different between the normal cervical tissue group and the CINI group,CIN?-?group,and cervical cancer group(?2=27.314,P<0.001;?2=32.117,P<0.001;?2=54.660,P<0.001).Correlation analysis showed that the correlation between Cdc2 and BAD in the apoptotic pathway in cervical cancer: Cdc2 and BAD positive expression were negatively correlated(r=-0.751,P<0.01);Cdc2 was positively correlated with HR-HPV typing(r=0.386,P<0.01);BAD was negatively correlated with HR-HPV typing(r=-0.445,P<0.01);Emi2 was positively correlated with HR-HPV typing(r=0.667,P<0.01).Conclusions Cdc2 and BAD can be regarded as reference standards for the degree of cervical tumor differentiation and clinical stage.The high expression of Emi2 gene promotes the malignant progress of tumor cells.Cdc2,BAD,and Emi2 play an important role in the occurrence and development of cervical cancer.The high expression of Cdc2 can lead to cell proliferation disorder,which is of great significance for the early diagnosis,invasion ability and malignancy of cervical cancer.The formation,treatment and prognosis of cervical cancer are of great significance;Emi2 has certain guiding significance in the diagnosis,degree of differentiation and malignant progression of cervical cancer.Cdc2,BAD and Emi2 are expected to become new targets for the diagnosis,treatment and prognosis of cervical cancer.In cervical cancer,the high expression of Cdc2 regulates the low transcription of BAD,which leads to the excessive proliferation of cervical cancer cells.The high expression of Emi2 can lead to cancerous transformation of cervical lesion cells.Figure 4;Table 20;Reference 131...
