Objectives To observe whether oral rehydration could improve the prognosis of sepsis mice by increasing the number of bone marrow cells and liver cells and promoting the transformation of bone marrow cells(BMCs)and liver cells to regulatory dendritic cells(CD11clowCD45RBhighDCs).Methods 1 Observe of the survival rate of thirty male septic mice which is healthy C57BL/6 mice,6-8 weeks old and weighing 20 ±1g.Cecal ligation and puncture(CLP)were used to establish a lethal sepsis model.These samples were randomly divided into 3 groups:sepsis group(C),low-dose oral rehydration group(T1),high-dose oral rehydration group(T2).In the group C,only lethal CLP was performed.After the mice aware from lethal CLP,group T1 and group T2 was administered 25ml/kg and 50ml/kg saline solution through mouth,respectively.All mice were feed in same environment with adequate water and food,and then observe the survival status and survival rate of the three groups of mice within 7 days.2 sixty healthy C57BL/6 mice,6-8 weeks old and weighing 20±lg,were selected and perform non-lethal CLP model.Mice were divided into 3 groups:group CLP,group L,group H.In the group CLP,only non-lethal CLP was performed.After the mice aware from non-lethal CLP,group L and group H was administered 25ml/kg and 50ml/kg saline solution through mouth,respectively.Five mice in each group died from cervical dislocation at 0,3,5,and 7 days,respectively.1)Observe the number of bone marrow cells and liver cells.Bone marrow cells and liver cells were taken from mice to made into cell suspensions.The number of bone marrow cells and liver cells were counted under the cell suspension microscope.2)Observe the number of CD11clowCD45RBhighDCs and the proportion of CD11clowCD45RBhighDCs/DCs.Dendritic cells(DCs)were purified from bone marrow and liver and then magnetically isolated with CD11clowCD45RBhighDCs.Flow cytometry instrument was used to testing the number of CD11clowCD45RBhighDCs and the percent of CD11clowCD45RBhighDCs/DCs in bone marrow and liver.Results 1 All mice appeared septic symptom,such as raised hair,accelerated heartbeat,decreased movement,increased secretions around the eyes,poor appetite,and poor drinking water.The survival rate of the group T2 was significantly higher than that in the group C(P<0.01),also higher than that in the group T1(P=0.012).The survival rate of T1 group was higher than that in group C(P=0.46).2 The number of bone marrow cells and liver cells:Compared with group CLP,bone marrow cells and liver cells increased in groups L and H.Compared with group L,bone marrow cells and liver cells increased in group H.1)The change of bone marrow cells:Compare with group CLP,bone marrow cells in group L increased significantly on the 7th day(P<0.05),H increased significantly on the 3th,5th and 7th day(P<0.05).Compare with group L,bone marrow cells in group H increased significantly on the 3th,5th and 7th day(P<0.05).2)Changes of liver cells:Compare with group CLP,liver cells in group L increased significantly on the 5th and 7th day(P<0.05),H increased significantly on the 3th,5th and 7th day(P<0.05).Compare with group L,liver cells in group H increased significantly on the 3th,5th and 7th day(P<0.05).3 The amount of CD11clowCD45RBhighDCs in bone marrow and liver:Compared with the group CLP,the amount of CD11clowCD45RBhighDCs in bone marrow and liver in group H and group L is increased.CD11clowCD45RBhighDCs in bone marrow and liver were increased in group H compared with group L.1)The number of CD11clowCD45RBhighDCs in bone marrow:Compare with group CLP,CD11clowCD45RBhighDCs in group L increased significantly on the 5th day(P<0.05),group H increased significantly on the 3th,5th and 7th day(P<0.05).Compare with group L,CD11clowCD45RBhighDCs in group H increased significantly on the 3th,5th and 7th day(P<0.05).2)The number of CD11clowCD45RBhighDCs in the liver:Compare with group CLP,CD11clowCD45RBhighDCs in group H increased significantly on the 3th,5th and 7th day(P<0.05).Compare with group L,CD11clowCD45RBhighDCs in group H increased significantly on the 3 th,5 th and 7 th day(P<0.05).4 the percent of CD11clowCD45RBhighDCs/DCs in the bone marrow and liver:Compared with the CLP group of mice,group H and L CD11clowCD45RBhighDCs/DCs in group H and L was increased.Compared with group L,the percent of CD11clowCD45RBhighDCs/DCs of bone marrow and liver in group H was increased.1)The percent of CD11clowCD45RBhighDCs/DCs in bone marrow:Compare with group CLP,the proportion in group L increased significantly on the 7th day(P<0.05),group H increased significantly on the 3th,5th and 7th day(P<0.05).Compare with group L,the proportion in group H increased significantly on the 3th,5th and 7th day(P<0.05).2)The percent of CD11clowCD45RBhighDCs/DCs in liver:Compare with group CLP,the proportion in group H increased significantly on the 3th,5th and 7th day(P<0.05).Compare with group L,the proportion in group H increased significantly on the 3th,5th and 7th day(P<0.05).Conclusions 1 Oral rehydration can significantly improve survival rate of sepsis mice.High dose(50ml/kg)oral rehydration has a higher survival rate than low dose(25ml/kg)oral rehydration.2 Oral rehydration can stimulate the proliferation of bone marrow cells and liver cells in sepsis mice,and increase the number and proportion of CD11clowCD45RBhighDCs in bone marrow and liver,with the highest effect in high dose(50ml/kg)oral rehydration group.3 Oral rehydration may compensate septic mice for impaired immunity and eradicate bacteria by mobilizing bone marrow cells and liver cells to differentiate into CD11clowCD45RBhighDCs.Figure 7;Table 5;Reference 136...
