Objective:To investigate the effect of CSF containing Huanglian Jiedu Decoction on the apoptosis and inflammation of BV2 cells induced by oligomer Aβ1-42 and its mechanism.Methods:BV2 cells were induced by oligomer Aβ1-42 and cell damage modelwas established.BV2 cells in logarithmic phase were randomly divided into blank control group,Aβmodel group,huperzine Acontaining CSF group,Huang Lian Jiedu deco-ction containing CSF low,medium and high dose groups.10%blank cereb-rospinal fluid,10%huperzine A containing cerebrospinal fluid,5%,10%and 20%Huanglian Jiedu decoction containing cerebrospinal fluid were added respect-tively.The apop-tosis of BV2 cells was detected by cell loss assay,the expression of IL-10,IL-6,IFN‐γin the supernatant of BV2 cells was detected by ELISA,and the expression of Arg-1and i NOS in BV2 cells was detected by Western blot.Results:1.Compared with the blank group,the apoptosis rate of BV2 cells in the Aβmodel group increased significantly(*P<0.05),the IL-10 in the supernatant decreased significantly(*P<0.01),and the levels of IL-6 and IFN‐γincreased significantly(*P<0.05).of BV2 cells induced by oligomer Aβ1-42.anti-inflammatory factor IL-10(*P<0.05)and decrease the level of pro-inflammatory factor IL-6 and IFN‐γin the supernatant of BV2 cells(*P<0.05).expression level of Arg-1 protein and decrease the expression level of i NOS protein in BV2 cells(*P<0.05).Conclusion:1.Oligomer Aβ1-42 can induce apoptosis and inhibit M2 type activation of BV2 cells.It can promote the activation of pro-inflammatory M1 type of BV2 cells,and release proinflammatory factors such as IL-6,IFN‐γ.2.The CSF containing Huanglian Jiedu Decoction can improve the release of inflammatory factors of microglia induced by oligomer aβ1-42,and its mechanism may be the same as that of Huanglian Jiedu Decoction,which can protect microglia,reduce microglia apoptosis,reduce IFN‐γsynthesis,promote the release of anti-inflammatory factor IL-10,and promote BV2 The activation of M2,the decrease of M1 and the decrease of IL-6 expression are related.
