| Objective: Based on the theory of "reduce the excess",high-fat diet-induced obesity(DIO)rats were used as the research carrier in this study.It aimed to observe the influence of pestle needle on body weight and irisin in DIO rats,and to explore the mechanism of improving obesity by means of pestle needle,so as to provide objective basis for clinical treatment of obesity.Methods: 1.40 1-month old SPF grade SD rats,male,weighing 70±5g,were randomly divided into 2 groups according to the random number table method after 3 days of adaptive feeding of common feed,including 8 rats in the control group and 32 rats in the high-fat group.The control group was fed with common feed while the high-fat group was fed with high-fat feed.Their length and weight were measured every week,and after 9 weeks,rats in the high-fat group who weighed more than 15% of the control group(6 rats)were selected as high-fat diet-induced obesity(DIO)rats.The final body mass,body mass increment and Lee’s index of each group were measured to comprehensively evaluate the establishment of the obese rat animal models.2.20 DIO rats were randomly divided into 3 groups: model group(6),exercise group(6)and pestle needle group(8).Meanwhile,6 rats fed with normal diet were used as the control group.Measure body weight and length every week,record general condition observation table.3.The body length and weight of rats in each group were measured and recorded respectively under fasting state on the ninth weekend.At the beginning of the 10 th week,the blank control group rats continued to be fed with ordinary feed without any other treatment.The model group rats continued to be fed with high-fat feed without any other treatment.Rats in the pestle needle group continued to be fed with high-fat feed and acupoint pressure;rats in the exercise group continued to be fed with high-fat feed and were swimming.After 22 days of continuous intervention,body length and weight were measured,and obesity rate,body mass,body mass increment and Lee’s index were calculated.Blood was taken from the femoral artery,serum was separated,GLU,TC,HDL-C,LDL-C and TG were detected,and serum irisin content was detected by ELISA;gastrocnemius muscle was taken,irisin m RNA was detected by RT-PCR,and FNDC5 protein expression level was detected by immunohistochemical method.Results: 1.Compared with the model group,the body mass of the pestle group and the exercise group was reduced after treatment,and the difference was statistically significant(P<0.05),while the difference between the pestle group and the exercise group was not statistically significant(P>0.05).2.After intervention,compared with the blank control group,TG value of the model group was higher,and the difference was statistically significant(P<0.01);compared with the model group,TG value of the pestle needle group was lower,and the difference was statistically significant(P<0.01);compared with the model group,TG value of the exercise group was lower,and the difference was statistically significant(P<0.05);while the difference between the exercise group and the pestle needle group was no statistical difference(P>0.05).There was no statistical difference(P>0.05)in GLU,TC,HDL-C and LDL-C among the groups.3.Compared with the model group,the serum irisin content of the pestle needle group was higher,and the difference was statistically significant(P<0.05).While there was no statistically significant difference between the remaining groups(P>0.05).There was no statistical difference in the content of gastrocnemius muscle irisin m RNA among the groups(P>0.05).Compared with the blank control group,the content of FNDC5 protein in the gastrocnemius tissue of the model group was significantly lower(P<0.01).Compared with the model group,the content of FNDC5 protein in the gastrocnemius tissue of rats in the exercise group was higher,and the difference was statistically significant(P<0.05).There was no significant difference between the other groups(P>0.05).Conclusions: 1.Pestle needle acupoint pressure can reduce the body weight and triglyceride levels of high-fat diet-induced obese rats.2.Pestle needle acupoint pressure can achieve weight loss by causing the increase of serum irisin in high-fat induced obese rats,which may be one of the mechanisms of weight loss by pestle needle. |
