Objective By observing the expression of articular cartilage and type II collagen in experimental rabbits with knee osteoarthritis,the therapeutic effect of human umbilical cord mesenchymal stem cells on knee osteoarthritis and the effect of ultrashort wave on this effect were clarified,which provided experimental and theoretical support for clinical application of umbilical cord mesenchymal stem cells combined with ultrashort wave in the treatment of knee osteoarthritis.Methods 26 male 6-month-old healthy New Zealand white rabbits were used to make the right hindlimb knee joint model by the modified Hulth method.7 weeks after the model was made,two of the experimental rabbits were randomly selected and killed by air embolism method.The gross and histological specimens were observed to verify the success of the model.After the model was established successfully,the remaining 24 experimental rabbits were divided into 4 groups by using the random number table method,namely,model group,ultrashort wave group,stem cell group and comprehensive group,6 in each group.The model group was free to move in the cage without intervention;the ultrashort wave group was intervened with ultrashort wave at the same dose at a fixed time every day;the stem cell group was intervened with the injection of HUC MSCs into the articular cavity;the comprehensive group was intervened with ultrashort wave after the injection of HUC MSCs into the articular cavity.After the intervention,the experimental rabbits in each group were killed.After the knee joint was cut,the articular surface and surrounding tissues were observed with naked eyes.The medial condylar cartilage of femur was taken,pathological sections and hematoxylin eosin(he)staining were performed,and the cartilage morphology was scored and statistically analyzed according to the modified mankins scoring criteria.The tibial plateau cartilage was taken and the soft tissue of each experimental rabbit was detected by real-time quantitative PCR The content of type II collagen in bone tissue was analyzed statistically.Results 1.Compared with model group,ultrashort wave treatment group,stem cell treatment group and comprehensive treatment group,mankins score of HE staining ofmedial condylar cartilage of femur decreased in turn,with statistical significance.2.Compared with model group,ultrashort wave treatment group,stem cell treatment group and comprehensive treatment group,the content of type II collagen in cartilage tissue increased in turn,and the results were statistically significant.Conclutions human umbilical cord mesenchymal stem cells and ultrashort wave have therapeutic effects on OA of rabbit knee,which can delay or block the progression of OA,reduce inflammatory reaction,repair articular cartilage damage and improve joint function.After the treatment of human umbilical cord mesenchymal stem cells combined with ultrashort wave,the therapeutic effect of ultrashort wave is better than that of human mesenchymal stem cells and ultrashort wave alone in the aspects of the gross appearance of the specimen,the Mankin's score under the cartilage light microscope and the real-time quantitative PCR content of type II collagen.Inflammation has a promoting effect.
