Chronic pain affects more than 30% of the population in China,and its treatment is still a difficult problem,so the discovery of new painkillers is increasingly urgent.Evodiamine(Evo)and rutaecarpine(Rut)are two major active components of Evodia rutaecarpa,a chinese traditional medicine which has been used as an analgesic for long time.However,their effects on mechanical hyperalgesia have not been evaluated.Our molecular simulation experiments showed that Evo and Rut docked to transient receptor potential cation channel subfamily V member 1(TRPV1),which was similar as capsaicin,and electrophysiological recording experiments confirmed that application of Evo(10 ?M)and Rut(50 ?M)activated TRPV1 expressed on HEK293 cell.Behaviorally,the application(i.p.)of Evo and Rut reduced mechanical hyperalgesia at a dose dependent manner,and this effect was blocked by capsazepine,a selective inhibitor of TRPV1.Furthermore,both Evo and Rut increased the time in open arms in elevated plus maze spent by mice with nerve injury.Therefore,these observations suggested that Evo and Rut reduced mechanical hyperalgesia and anxiety in mouse with nerve injury or inflammation via TRPV1.
