Fibrosarcoma is a rare malignant tumor originating from mesenchymal tissue.It has high invasiveness,and can metastasize to the lung,skeletal system,liver and other parts of the body through the lymphatic system or blood.Compared with other soft tissue sarcomas,fibrosarcoma has a higher recurrence rate and a worse prognosis.Surgical resection is the main treatment for fibrosarcoma,but this method is not suitable for tumors located in deep tissue.In addition,the lack of specific biomarkers makes it difficult to diagnose fibrosarcoma accurately.Therefore,looking for molecular probes that can specifically recognize fibrosarcoma is of great significance for early diagnosis,targeted therapy and accurate prognosis of tumors.As a probe molecule,aptamers are widely used in biomedicine,biochemistry and other fields because of their high affinity,high specificity and low immunogenicity.In this work,we found that DNA aptamer S11 e could recognize fibrosarcoma cell HT1080 by non-SELEX method,but could not recognize human embryonic lung fibroblast MRC-5.S11 e has good binding affinity with HT1080 cells,and the equilibrium dissociation constant is in the range of nanomole.In vivo and in vitro fluorescence imaging further confirmed that S11 e recognized tumors in HT1080 xenograft nude mice model and fibrosarcoma clinical tissue.In addition,S11 e could be internalized into HT1080 cells under physiological conditions and bound to mitochondria by confocal laser irradiation.CCK-8 toxicity test and Annexin V-FITC flow cytometry showed that S11 e has biological function on HT1080 cells and can promote cell apoptosis.Scratch test showed that S11 e could inhibit the migration of HT1080 cells.These results indicate that S11 e can not only target HT1080 cells,but also inhibit HT1080 cells from cancer.It may be used as a molecular drug in early diagnosis,targeted therapy and prognosis of fibrosarcoma.
