German chamomile?Matricaria chamomilla L.?is an ancient aromatic plant cultivated in large areas in China.In recent years,the research on the activity of German chamomile essential oil has focused on the biological activities such as antibacterial,anti-allergic and anti-inflammatory.In the field of wound healing,it has been used as folk medicine in wound healing treatment,but the mechanism of its effect on skin wound healing has not been explored.This paper is aimed to investigate the effects of German chamomile essential oil on skin wound healing and related mechanisms,in order to provide a scientific basis for the application of German chamomile essential oil in skin wound healing.The main results of this paper are as follows:1.The effects of German chamomile essential oil on the proliferation of skin fibroblasts were determined by CCK-8?Cell Counting Kit-8?method.The effects of German chamomile oil on the migration of fibroblasts were detected by scratch method.The results showed that German chamomile essential oil significantly promoted the proliferation and migration of fibroblasts in a dose-dependent manner.Its ability to promote the proliferation of skin fibroblasts was weaker than that of lavender and mandarin essential oils,and its ability to promote the migration of skin fibroblasts was stronger than that of lavender and mandarin essential oils.2.The skin toxicities of German chamomile essential oil with content???of 2%,5%and 8%were detected respectively by the acute skin toxicity test of mouse.As a result,no obvious toxicity was observed.The excision wound model was established to evaluate the effects of German chamomile essential oil with concentration???of 2%,5%and 8%on the wound healing in mouse.The results showed that low,medium and high concentrations of German chamomile essential oils significantly improved the wound healing rate of the skin of mouse,increased the number of fibroblasts and the thickness of dermis of the wound skin,and promoted the formation of the skin accessory organs.The medium concentration of German chamomile essential oil has the best promoting effect.3.The excision wound model was used to further explore the mechanism of German chamomile essential oil promoting wound healing in mice.The contents of hydroxyproline,superoxide dismutase,basic fibroblast growth factor and transforming growth factor?1 in the wound skin were determined by this model.The collagen synthesis in the wound skin was analyzed by collagen fiber staining.The results showed that German chamomile essential oil up-regulated the secretion and expression of cytokines basic fibroblast growth factor and transforming growth factor?1,increased superoxide dismutase activity,promoted the proliferation and migration of skin fibroblasts,promoted the extracellular matrix of collagen synthesis by skin fibroblasts,reduced the oxidative stress response of wound tissue and promotes angiogenesis,and finally made the tissue exhibit better remodeling.
