Objective: To establish a high-sugar and high-fat diet for 8 weeks combined with intraperitoneal injection of small-dose streptozotocin(STZ)to establish a rat model of type 2 diabetes.Effects of successful model-building on the expression of TGF-?1,MMP-9 / TIMP-1 protein in kidney of type 2 diabetic rats.Research methods: Forty male SD rats were randomly divided into a quiet control group(NC),a diabetic model(DM),a medium-intensity exercise group(MICT),and a high-intensity intermittent exercise group(HIIT),with 10 in each group.The NC group was fed with ordinary feed,and the DM group was fed with high-sugar and high-fat feed,and they were free to move in cages.The MICT and HIIT groups were fed with high-sugar and high-fat.At the end of the 8th week,after measuring the weight and fasting blood glucose(FBG)of rats in each group,fasting for 12 hours overnight,the NC group was injected intraperitoneally with citric acid buffer,and the DM,MICT,and HIIT groups were administered once every 30 mg / kg of body weight.Intraperitoneal injection of 2% STZ solution,on the 7th day of STZ injection,the weight,random blood glucose,and orbital blood of rats in each group were measured,and serum transforming growth factor(TGF-?1)was measured by ELISA in each group.The kidney was weighed after removal of the capsule,and a part of the cortex of the left kidney was frozen and stored at-80 ° C to detect protein.The Western Blot(WB)method was used to detect renal tissue TGF-?1,MMP-9 and TIMP-1.Results: 1.There was no statistical difference in the initial weight of each group(P> 0.05).After 8 weeks of high-sugar and high-fat diet,there was no statistical difference in body weight between the NC group and the DM,MICT,and HIIT groups,but MICT,Compared with the DM group,the body weight of the HIIT group showed a downward trend.2.There was no statistical difference in serum TGF-?1 between NC,DM,and MICT groups.Compared with DM and MICT groups,there was a statistically significant difference in HIIT group(P <0.05),which was significantly increased.3.Renal tissue TGF-?1,DM group was significantly higher than NC group(P< 0.05),there was no statistical difference between NC,MICT and HIIT groups.4.MMP-9 and TIMP-1 protein in kidney tissue: MMP-9 has no statistical difference in the kidneys of each group.TIMP-1 content in the DM and MICT groups is significantly higher than that in the NC group(P< 0.05).There is no statistics in the HIIT group compared with the NC group difference.Research conclusions: 1.Different types of exercise interventions have a tendency to reduce the weight of diabetic rats.There is no significant difference in the ability of two exercise methods to reduce the weight of rats.2.During the construction of T2 DM rat model with 8 weeks of high-sugar and high-fat feeding combined with low-dose STZ,the changes of plasma TGF-?1 and kidney TGF-?1 were in reverse trend.However,high-intensity intermittent exercise can make the plasma TGF-?1 level in rats significantly higher than those in the MICT and DM groups.3.Both high-intensity intermittent and moderate-intensity exercise can delay the overexpression of TGF-?1 in renal cortical tissue.4.Compared with moderate-intensity exercise,high-intensity intermittent exercise can also delay the time of excessive expression of TIMP-1 protein,thereby delaying the progression of renal fibrosis.
