Skin is the largest organ of human body,which plays an important role.In real life,various kinds of skin injuries inevitably occur,and large-scale skin repair is also one of the clinical difficulties.This study attempts to apply bio-3D printing technology to the construction of artificial skin and achieve the purpose of repairing and reconstructing skin tissues.By re-evaluating and calibrating the existing bio-3D printing system in the laboratory,the effects of relevant control parameters(such as flow rate,voltage frequency,nozzle diameter,working distance,substrate speed,etc.)on the printing process of bio-materials were systematically studied,and the printing and forming of bio-active materials represented by cells were attempted by this technology.The results show that the technology can successfully encapsulate cells in hydrogel microspheres and maintain high activity under optimized conditions.Through adjusting these parameters to balance the forces exerted on the microspheres during the printing process,the diameter of the microspheres can be controlled in the range of 100-600 microns.This 3D printing technology may be able to extend the potential use of bioactive materials such as microspheres to other applications,such as cancer biology and drug screening,and may also serve as an effective platform for exploring their applications in tissue engineering.In addition,an artificial skin model was constructed using our bio-3D printing system in our work.Through various physical and chemical characterization,and in vivo and in vitro experiments,it is proved that the model not only has good biological activity,strong ability of inducing cell growth,strong recruitment effect on cell migration and up-regulation of gene expression of related factors,but also has a good effect on wound healing in vivo animals,and shows a strong effect in reconstruction of skin tissues!...
