Objectives:Obesity has become a globally public health problem.It has become a great challenge for relevant researchers and clinical workers to clarify the pathogenesis so as to diagnose and correct obesity.MicroRNAs(miRNAs)are small molecules of nucleic acid with a length of 18-22 base Numbers,which can regulate the transcriptional level of post-transcriptional mRNA.More and more studies have shown that such molecules are closely related to metabolic diseases such as obesity.This study intends to research the correlation between miRNAs in the circulating serum of obese patients and obesity,and preliminarily explore the possible mechanism of abnormally expressed miRNAs in obesity,so as to provide new ideas for clinical diagnosis and treatment of obesity.Methods: Firstly,the clinical samples were divided into normal weight and obesity groups by body mass index,and the expression of miRNAs in serum was detected by real-time quantitative PCR technology,and the differentially expressed miRNAs were screened.Secondly,the adipose-related precursors were used as the in vitro experimental models to detect the effects of miRNAs,and used the above models to observe the differentiation process of white and brown adipocytes,as well as the RT-qPCR technology and protein level detection and other molecular biological detection of the important genes were used to research adipocyte differentiation.Next,bioinformatics and related experiments were applied to predict and confirm the target genes that miRNAs might act on the process of adipocyte differentiation.Results: After the detection of miRNAs in serum samples,it was found that miR-129-5p increased with weight gain.In the adipogenic differentiation experiment,it was found that miR-129-5p was involved in the adipocyte differentiation process,and the expression of the signaling regulators of C/EBP? and PPAR?2,the key factors related to lipid differentiation was decreased.Protein markers including UCP1 and CIDEA were decreased in mature adipocytes.Thus,it was preliminarily inferred that the miRNA inhibited adipocyte differentiation.miR-129-5p down-regulated ATG7 gene and mediated autophagy signaling pathway,thereby inhibiting adipocyte differentiation.Conclusion: 1.miR-129-5p may inhibit adipocyte differentiation by mediating autophagy signal pathway.2.The expression of miR-129-5p in serum samples may be a biomarker for predicting obesity and other metabolic diseases.
