PurposeTo established an animal model of atherosclerosis in type 2 diabetes,and to further study of to further study the effect of APN on pathophysiological changes after angioplasty,and explored the role of APN in protecting endothelial cells,promoting re-endothelialization,and reducing restenosis,to analyse the feasibility and molecular mechanism of apply APN in reducing restenosis after angioplasty.Methods 1,Model and group establishmentAll of the animals to established atherosclerosis model,and then,PTA was performed on the lesions formed by AS,which were randomly divided into APN experimental,group(Test Group)and control group.The test Group group was supplemented with adiponectin,and the control group was injected with normal saline,once every other day for 3 times.On day 70,angiographic examination of the two groups of animals was performed to understand the vascular patency and stenosis,blood and the iliac-femoral artery specimens were obtained for pathological study and molecular biological analysis.2,The test of serum indicatorsTotal cholesterol(TC),triglyceride(TG),low density 1ipoprotein cholesterin(LDL),high density lipoprotein cholesterol(HDL),nitric oxide(nitric)Oxide,NO)and APN.3,Vascular histopathology molecular biology testEvans blue staining evaluate the endometrialization.The general morphological changes of the vessel wall were assess by Hatmatoxylin-Eosin staining.Mavot staining analysis foam cell distribution,vascular smooth muscle cells and collagen hyperplasia,PCNA immunohistochemical analysis cells proliferation.Oil red O staining was used to found lipid deposition,immunofluorescence was used to evaluate the damage of reactive oxygen species and mitochondrial DNA,and Western blotting was used to learn intracellular cytoplasmic cytochrome C content.Image analysis was performed using Image-Pro Plus 6.0,and statistical analysis was performed on spss 25.0.ResultsThe blood glucose,TC,TG and LDL-C in the control group were higher than test group(P<0.05).There was no significant change in HDL-C in the short term,no significant difference in two groups(P>0.05).Test group APN and NO concentration were higher than the control group(P<0.05).The Ewan's blue staining showed that the test group had a larger endometrial area than the control group(P<0.05),and the consequent significant difference.Digital subtraction angiography images was analyzed the vascular stenosis.The restenosis in test group was smaller than in the control group(P<0.05).The ROS and MtDNA fluorescence analysis showed that the ROS content and MtDNA damage in the test group were less than the control group(P<0.05),and the lipid content in the oil red O staining test group was less than the control group(P<0.05).Western-Blot detection of Cyt-C showed that the test group was smaller than the control group(P<0.05),indicating that the ROS accumulation and mitochondrial damage in the test group were less than the control group.HE staining shows the thickness of the intima of the test group was significantly thicker than control group.A large number of foam cells were seen in the control group intima.The intima-media thickness(IMT)ratio,intimal area,and intima collagen were analyzed by Mavot staining,the fibrous tissue and proliferating cell nuclear in the test group was less than the control group(P<0.05).ConclusionEndothelial injury caused by angioplasty is one of the important causes of restenosis.APN can regulate the metabolism and protect the endothelial cells,promote re-intimalization to reduce the occurrence of postoperative vascular restenosis,and positive remodeling of blood vessels.
