| Objective: By observing the effect of Didang Decoction on the expression of inflammatory factors in the inflammation model of BV-2 cells,to study the Didang Decoction’s molecular mechanisms of the treatment of inflammatory reaction after cerebral hemorrhage,and determine it’s therapeutic targets.Methods: 1.Screening drug concentration: BV-2 cells were cultured with complete medium that containing 0.8,4,20,100,500,1000 μg/mL of the Didang Decoction for 24 hours,and the effects of different concentrations of Didang Decoction on BV-2 cells’ survival rate were detected by MTT assay;2.The effect of Didang Decoction on the TNF-α,IL-1β,IL-6 mRNA in BV-2 cell inflammation model was detected by Real Time PCR;3.ELISA method was used to detect the effect of Didang Decoction on the expression of TNF-α in the supernatant of the inflammation model of BV-2 cells;4.Western Blot method was used to detect the effect of Didang Decoction on the expression of IL-1βin the inflammation model of BV-2 cells.Results: 1.After co-culture of BV-2 cells with different concentrations of Didang Decoction for 24 h,the group of 0.8,4,20,100μg/mL of Didang Decoction was not inhibited cell viability,which can be used in subsequent experiments;2.The expression of TNF-α,IL-6 and IL-1βmRNA were significantly increased at the inflammation model of BV-2 cells,and the groups of witch contain 4,20,100 μg/mL of Didang Decoctioncan effectively reduced the expression of TNF-α and IL-1β mRNA,and there was a quantitative dependence.However,there was no significant inhibition on the expression of IL-6 mRNA.3.The content of TNF-αin the supernatant of the inflammation model of BV-2 cells was significantly increased,and the concentration was decreased in the group of 4,20,100μg/mL Didang Decoction;4.The expression of IL-1β in the inflammation model of BV-2cells increased significantly,and the expression of IL-1β decreased by 4,20,100 μg/mL Didang Decoction,which was quantitatively dependent.Conclusion: Didang Decoction has no obvious effect on the expression of IL-6 in BV-2 cell inflammation model,but it can effectively inhibit TNF-α and IL-1β.These may be the molecular mechanism of Didang Decoction’s treatment of inflammatory reaction aftercerebral hemorrhage. |
