Objective:To observe single-nucleotide polymorphisms(SNPs)at six sites in the 1,4,5-triphosphate inositol 3-kinase C(ITPKC)and phospholipase C-like 1 protein(PLCL1)genes and the Han population in Ningxia and the correlation between stable COPD and related pH.Methods:250 cases of Han patients with stable COPD who met the research objectives of Ningxia Hui Autonomous People's Hospital were randomly selected in this experiment,including 103 patients with COPD-related PH and 147 patients with COPD non-PH,and selected a total of 127 healthy controls of the Han nationality,and the ITPKC loci rs2288450,rs10411159,rs2290692,rs1045705,rs17713068,and PLCL1 locus rs9789480 genotype were detected using Agena MassARRAY.The statistical analysis was performed using SPSS 19.0 statistics software to compare each differences in group allele and genotype frequencies.Results:? There was no significant difference in age,gender and smoking index between the COPD group and the control group,the COPD non-PH group and the COPD-PH group.?The average FEV1/FVC(78.54±10.47)in the control group was higher than that in the COPD group(50.94±9.96),and the average PASP(21.68±2.38)was lower than that in the COPD group(56.88±10.56).The average FEV1/FVC(63.42±4.53)of the COPD non-PH group was higher than that of the COPD-PH group(51.46±3.06),and the average PASP(31.84±7.00)was lower than that of the COPD-PH group(58.67±9.77).?Sites rs10411159 and rs1045705 did not meet the Hardy-Weinberg equilibrium test.The genotype frequencies of rs2288450,rs2290692,rs17713068 and rs9789480 all accorded with Hardy-Weinberg equilibrium.?The alleles and genotype frequencies of rs2288450 and rs9789480 loci were significantly different between the COPD group and the control group(P<0.05).The rs2288450 locus was compared with COPD patients carrying the CC genotype and C allele.the prevalence of carrying CT and TT genotypes was 0.55 and 0.32 times that of carrying CC genotype,and the prevalence of carrying T allele was 0.54 times that of carrying C allele,suggesting that CT,TT genotype,T Alleles may be protective factors for COPD patients,which can reduce the prevalence of COPD.Compared with patients with COPD carrying the CC genotype and C allele at the rs9789480 locus,the prevalence of carrying CA and AA genotypes was 0.41 and 0.20 times that of carrying the CC genotype.The prevalence of carrying the A allele was Carrying the C allele 0.40 times,suggesting that CA,AA genotypes and A alleles may be protective factors of COPD,which can reduce the prevalence of COPD.?The allele and genotype frequency distribution of rs9789480 locus was statistically significant between the non-PH group and the COPD-PH group(P<0.05),which was similar to that of patients with COPD-PH who carried the CC genotype and C allele.The prevalence of CA and AA genotypes was 0.07 and 0.03 times that of CC genotypes,and the prevalence of A alleles was 0.10 times that of C alleles,suggesting that CC genotypes and C Alleles are risk genes that increase the prevalence of COPD-PH.?There was no statistically significant difference in the distribution of alleles and genotype frequencies in rs2290692 and rs 17713068 loci between the control group and the COPD group(P>0.05).There were no statistically significant differences in the frequency of alleles and genotypes of rs2290692,rs17713068,and rs2288450 loci between the COPD-related PH group and the COPD non-PH group(P>0.05).Conclusion:The rs2288450 locus of ITPKC gene and the rs9789480 locus of PLCL1 gene may be related to the susceptibility to COPD and COPD-PH.The rs2288450 locus carrying the CA,AA genotype and A allele may be a protective factor for COPD The prevalence of COPD;carrying the CA,AA genotype,and A allele at rs9789480 may be protective factors for COPD and COPD-PH,which can reduce the prevalence of COPD and COPD-PH.
