Preliminary Study On The Diagnostic Values Of IMP-3? PAX-8?Galectin-3 In Well-differentiated Thyroid Carcinoma

Objective:To investigate the significance of differential diagnosis of insulin-like growth factor mRNA binding protein 3(IMP-3),paired-box gene 8(PAX-8)and galectin-3(Galectin-3)in the pathology of well-differentiated thyroid carcinoma.Methods:We collected 165 cases of thyroid lesions from the files of First Hospital of Shanxi Medical University from March to December 2018.PTCs included 57 cases of classic papillary carcinoma(CLPTC)and 18 cases of follicular papillary carcinoma(FVPTC).And there were 18 cases of follicular thyroid carcinoma(FTC).There were72 cases of benign thyroid nodules(BTN),including 52 cases of nodular goiter(NG)and 20 cases of follicular adenoma(FA).All samples were fixed in formalin and embedded in paraffin,and 4 ?m serial sections were cut continuously.The immunohistochemistry(IHC)staining was used to detect the expression of IMP3,PAX8 and Gal-3 in the thyroid lesions.All specimens were double-blinded by two physicians without knowing the diagnosis of the original case.SPSS 22.0 statistical software was used to analyze the data using non-parametric tests and rank tests.Based on the IHC score ? 3 +,the sensitivity(SE),specificity(SP),positive predictive value(PPV),and negative predictive value(NPV)of three marker factors to distinguish PTC and BTN,FTC and FA,and FVPTC and FA were counted respectively(NPV)and diagnostic accuracy(AC).Results:IMP3,PAX8 and Gal-3 expressed in all the five groups.The expression of the three markers PTC and BTN were statistically different(P <0.05).The expression of IMP3 in FTC was compared with FA.Statistical difference(P <0.05);The expression of IMP3 and Gal-3 in FVPTC was statistically different from FA(P <0.05).In the case of PTC and BTN,FTC and FA,FVPTC and FA differential diagnosis,the SP,PPV and AC of IMP3 were the highest.Conclusion:IMP-3 and Galectin-3 have potential diagnostic values in comparing PTC with BTN and show highly diagnostic values to identify follicular lesions.PAX8 requires more consideration of the intensity and distribution of staining when separately distinguishing highly differentiated thyroid cancer and benign lesions.
