Objective:RNA m6 A methylation regulators can dynamically regulate tumorigenesis and progression.But the role of RNA m6 A methylation in kidney renal clear cell carcinoma is not clear.The purpose of this study is to analyze the characteristics of RNA m6 A methylation in kidney renal clear cell carcinoma and its correlation with clinical characteristics and prognosis.Methods:In this study,we analyzed the correlation between the 23 RNA m6 A regulatory factors and clinical characteristics and prognosis in 611 kidney renal clear cell carcinoma samples existing in The Cancer Genome Atlas database.The difference of mRNA expression levels of regulatory factors in tumor and paracancer normal tissues of kidney renal clear cell carcinoma was examined by Wilcox rank test.The correlation between the mRNA expression of methylation regulatory factors in tumor tissue was analyzed,and Pearson correlation coefficient was calculated.Consensuscluster analysis was used to determine whether the mRNA expression of regulatory factors in tumor tissues could classify tumor.Principal component analysis was used to determine whether the mRNA expression of regulatory factors can represent the expression characteristics of tumor tissues.To analyze the relationship between tumor classification and prognosis,KMsurvival analysis was used to draw the survival curve,and log rank test was carried out.To analyze the correlation between tumor classification and clinical characteristics,and to compare the differences of clinical characteristics by Chi square test.The correlation between the mRNA expression of regulatory factors and prognosis in tumor tissue was analyzed.First,single factor Cox analysis was used to identify the genes related to prognosis.Then the Cox proportional hazards model was constructed by Lasso regression,and the risk score was calculated.The median of the risk score was taken as the cut-off value,and the patients were divided into high-and low-risk group.Compare overall survival between high and low risk groups by KM analysis.The prediction value of lasso regression Cox proportional risk model was evaluated by area under curve of operating characteristic curve.Chi square test was used to evaluate the difference of clinical characteristics between high and low risk groups.Single factor and multi factor Cox analysis were conducted to select independent predictors of overall survival.Results:In this study,we found that the mRNA expression of multiple RNA m6 A methylation regulators was different between paracancerous normal and tumor tissues of kidney renal clear cell carcinoma.There was a certain correlation between the mRNA expression of RNA m6 A methylation regulators in kidney renal clear cell carcinoma.Consensuscluster analysis showed that the tumor could be classified according to the mRNA expression of RNA m6 A methylation regulatory factors in kidney renal clear cell carcinoma,and the clusters were related to clinicalpathological characteristics and prognosis.The risk model based on the mRNA expression of eight RNA m6 A methylation regulators in kidney renal clear cell carcinoma had a good predictive value,was related to clinical characteristics,and was an independent risk factor for overall survival.Conclusion:The mRNA expression of RNA m6 A methylation regulators in kidney renal clear cell carcinoma are related to clinical characteristics.A risk model based on the mRNA expression of eight RNA m6 A methylation regulators can be used to predict the overall survival rate of patients with kidney renal clear cell carcinoma.
