Livin Promotes Differentiation Of Peripheral Blood CD4~+T Cells To Th2 By Binding To GATA3 And Participates In The Development Of Airway Allergic Diseases

Objective:To investigate the interaction between livin and GATA3 in CD4~+T cells in peripheral blood of patients with airway allergic disease(AAD),and to clarify the mechanism of livin promoting the formation of Th2 differentiation advantage.Methods:30 AAD patients(AAD group)and 30 healthy people(control group)who were treated in our department from March 2017 to March 2019 were collected,and peripheral blood was collected from two groups.CD4~+T cells were obtained by magnetic bead sorting.real-time quantitative polymerase chain reaction is used to detect the expression levels of livin mRNA,IL-4 mRNA,and GATA3 mRNA in two groups,observe the levels of livin and GATA3,and observe the relationship to IL-4 Expression;Western blotting was used to detect the expression levels of livin and IL-4 in peripheral blood CD4~+T cells in two groups;Co-immunoprecipitation(Co-IP)was used to analyze whether there is interaction between livin and GATA3.Results:Compared with the control group,the expression levels of livinmRNA,IL-4mRNA,and GATA3mRNA in the AAD group were increased,and the difference was statistically significant(P<0.05).The expressions of livin and IL-4 were positively correlated(r=0.5616,P<0.01),meanwhile,the expressions of GATA3 and IL-4were positively correlated(r=0.678,P<0.001);Among the two groups of peripheral blood CD4~+T cells,the expression levels of livin and IL-4 in the AAD group were higher than those in the control group,and the difference was statistically significant(P<0.05);GATA3 was detected in the protein complex co-precipitated with livin antibody,and livin was also detected in the protein complex co-precipitated with GATA3 antibody.Conclusion:Livin and GATA3 interact in the peripheral blood CD4~+T cells of AAD patients.Livin and GATA3 form a complex to increase the expression of IL-4 to promote the differentiation of Th2 cells,thereby participating in the occurrence of airway allergies.
