Objective:To investigate the protective effect of lilarutide on cardiologic functions in type 2 diabetic rats and explore its mechanism,providing a new idea for the treatment of diabetic cardiomyopathy in the clinic.Methods:40 Six-week-old health male SD rats were randomly divided into two groups:control group and modeling group.The two groups rats were separately fed with normal diet and high-sugar and high-fat diet for four weeks.Then,the rats model of diabetic cardiomyopathy were induced by injecting of 40mg/kg STZ solution and fed with high sugar and high fat diet for one month.The DCM rats were randomly divided into three groups:model group,high dose lliraglutide group and low dose liraglutide group.The rats were subcutaneous injected with different dose of lliraglutide for 8 weeks,in which recored their weight and fasting blood glucose each week.At the end of the treatment,all the rats were weighed and anaesthetized to retained blood serum to test blood glucose and serum lipid,as well as removed the whole myocardial tissue and weigh it.HE stainning was used to observe morphological changes of cardiac tissues.Immunohistochemistry was used to detect the expression of PARP-1 in cardiac tissues.Results:1.General situation and changes of body weight and heart weight indexCompared with the control group,the rats in model group demonstrated symptoms such as polyuria,polydipsia,polyphagia,dry hair,and decreased activity,as well as mats were obviously moist.After intervention with different doses of liraglutide,the performance above has changed slightly,especially in the high-dose group.2.Blood gluose and serum lipidCompared with the control group,the model group exhibited higher FPG?TC?TG?LDL-C?Lp-PLA2,(P<0.05).While compared with model group,these blood biochemical indexs decreased significantly in the intervention group,the high-dose group was more obvious(P<0.05),but it was still higher than the control group.And there is no significant difference of HDL-C in all groups(P>0.05).3.Morphological changes of cardaic tissuesCompared with the control group,myocardial interstitial collagen fiberc is significantly increased,and the disorders of myocardial fibers were obvious in the DCM group.And the above-mentioned changes are significantly improved after injecting liraglutide,especially in high-dose group.4.The expression of PARP-1 in cardiac tissuesCompared with control group,the expressions of PARP-1 in modle group increased obviously,the difference was statistically significant(P<0.05).Compared with modle group,the expressions of PARP-1 in the two dose groups decreased obviously,especially in the high-dose group,but it didn't recover to normal level.The difference was statistically significant(P<0.05).Conclusion:1.Low-dose STZ combined with high-sugar and high-fat diet and continued feeding for 4 weeks has successfully established DCM rats.2.Liraglutide can improve body weight and heart weight index,as well as FPG?TC?TG?LDL-C?Lp-PLA2 levels in rats with type 2 diabetic cardiomyopathy,and its effect is dose-dependent.3.Liraglutide may decrease myocardial PARP-1 expression in type 2 diabetic cardiomyopathy rats,and play a protective role in myocardial injury in diabetic cardiomyopathy rats.
