Sodium Selenite Provides A Protective Effect For The Aortic Endothelial Damage Induced By Silver Nanoparticle Exposure

Objective The study aimed to investigate the potential adverse effects induced by silver nanoparticles(AgNP)exposure in terms of oxidative stress,and mitochondrial injury in aorta,and sodium selenite shows protection from AgNP-induced adverse effects.Methods Sprague Dawley rats were divided into 4 groups(n=8),named the groups as following: Control group A,AgNPtreated group B,Se treated group C,Se+ AgNP treated group D.For group B and D,the rats need intratracheal instillation(IT)with the AgNP.Group A and C received a similar volume of 0.9% NaCl IT.Group B and C were intraperitoneal injection(IP)with sodium selenite(0.2mg/Kg BW,7 days).Vascular pathological alterations and ultrastructural changes were assessed using hematoxylin and eosin(HE)staining and electronic microscopy.Collagen deposition in aorta was assayed by Masson's Trichrome Staining.The generation of reactive oxygen species(ROS)in aortawas detected by the fluorescent probe.The cleaved caspase and mitochondrial fission marker were measured by immunofluorescence and western blot,respectively.Results The HE staining outcomes did not showed obviously histopathologic changes.AgNP enhanced collagen deposition,damaged the endothelial cell of aorta,nucleus was exposed and AgNP was deposited in it.The mitochondria of endothelial cells were edama and the cristae were dissolved and disappeared.AgNP increased the generation of ROS,increased protein expression of Drp1 and Fis1,activated cleaved caspase 3.Treatment with sodium selenite alleviated AgNP-induces injuries as we described aboveConclusion It is concluded that AgNP damaged the endothelial cell of aorta by increasing oxidative stress,elevated mitochondrial fission and activated caspase 3.Treatment with sodium selenite is capable of preventing the above adverse effects induced by AgNP.
