Black bean(Glyeine Soja Sieb.et.Zucc)was widely distributed in Northeast China,Qingren is a kind of it.They are rich in protein and minerals.They have anti-aging,eye-catching and long-lasting effects,and are known as "black pearls".Qingren black bean skin are rich in Anthocyanin,which can scavenge free radicals in the body and has strong antioxidant effect.Anthocyanins have anti-oxidation,anti-cancer,promote fat metabolism,prevent cardiovascular diseases,and also have the effect of lowering blood sugar and protecting vision.In this paper,Qingren black bean skin was used as the research object.Qingren black bean skin was extracted with 95 % ethanol refluxed.1 kg of Qingren black bean skin was extracted with 10 L ethanol,and extracted three times,each time for two hours to obtain ethanol extract of Qingren black bean skin.The obtained extract was rotary evaporated by vacuum recovery at 60 ℃ to recover ethanol,and the ethanol concentrate of Qingren black bean skin was obtained,and then the concentrate was freeze-dried to obtain 99.88 g of dried extract of Qingren black bean skin ethanol extract(i.e.,crude extract of Qingren black bean skin Anthocyanin),and the extraction rate was 9.98 %.The crude extract of Qingren black bean skin was separated by extracting with ethyl acetate and extracting with n-butanol to obtain each extract.Extracts obtain from obtain were separated and purified by Thin-layer Chromatography and High Performance Liquid Chromatography.Extracts were analyzed by Liquid Chromatograph-mass Spectrometer.Four compounds isolated from the Qingren black bean skin were identified,which were Cyanidin Delphinidin-3-O-glucoside,Malvidin and Methyl Anthocyanin.In this paper,the antioxidant activity of Anthocyanins was tested in vitro and in vivo.In the free radical scavenging experiment of 1,1-diphenyl-2-picrylhydrazyl(DPPH)in vitro.DPPH free radical scavenging rate of ethanol extract average was 86.336 %.The DPPH free radical scavenging rate of ethyl acetate layer extract average was 80.798 %.DPPH radical scavenging rate of n-butanol extraction layer average was 72.424 %.The anti-oxidation test of Qingren black bean skin anthocyanin was carried out by testing the lipid oxidation product malondialdehyde(MAD)content,protein carbonyl content,and sample glutathione,in the liver tissue of mice fed with Anthocyanin.Compared with the control group,the content of MAD in lipid oxidation products decreased,the content of protein carbonyl was also significantly decreased,and the content of GSH in samples increased.The test mice were Kunming mice.The high concentration sample in the model group A was 1 mg/mL,and the low concentration sample in the B group was 0.2 mg/mL.The C group was the control group,and the control group was given distilled water.The experimental mice were intragastrically administered 0.5 mL/day,5 mice per group,and continuously administered for 30 days.The average lipid oxidation product of group A was 0.1322 mg/mL,the average lipid oxidation product of group B was 0.249 mg/mL,and the average lipid oxidation product of group C was 0.3124 mg/mL.The average protein carbonyl content of group A mice was 5.33 nmol/mg,the average protein carbonyl content of group B mice was 10.15 nmol/mg,and the average protein carbonyl content of group C mice was 16.90 nmol/mg.The average glutathione content of group A mice was 147.3 umol/g,the average glutathione content of group B mice was 90.7 umol/g,and the average glutathione content of group C mice was 58.0 Umol/g.All three results showed that Anthocyanins in Qingren black bean skin had a certain antioxidant activity.Based on the antioxidant activity of Anthocyanins,Anthocyanin was prepared from Qingren black bean skin,and Anthocyanin was optimized.Orthogonal experiment,Anthocyanin drink with good color and texture was obtained.The process flow was as follows: Qingren black bean skin solvent was extracted with 95 % ethanol refluxed.1 kg of Qingren black bean skin was extracted with 10 L ethanol,and extracted three times each time for two hours to obtain ethanol extract of Qingren black bean skin.Using orthogonal experiments to get the optimal solution is that dissoved 5 g extract with 1L water,then added VC 0.01 g,xylitol 0.5 g.Then filtered and embedmented to get the finished product.The finished product was obtained as a reddish brown transparent liquid.After evaluation by 10 people,the average score was 95 points.
