BackgroundLung cancer is one of the malignant tumors with the fastest growth in morbidity and mortality.It poses a great threat to people's health and life.LncRNA ATB plays an important role in the development of various malignancies by competitively binding to miR-200,thereby upregulating the expression of the ZEB1 and ZEB2 families,and thereby inducing epithelial-mesenchymal transition and cancer cell invasion.However,the function of ATB in lung cancer has not been studied.This study aimed to investigate the role of ATB in lung cancer.ObjctiveTo study the effect of lncRNA ATB on lung cancer proliferation and metastasis.MethodsIn order to elucidate the role of ATB in lung cancer,reverse transcription? quantitative polymerase chain reaction was used to detect the expression of ATB in tumor tissues and corresponding non? tumor lung tissues from 32 patients with lung cancer.Furthermore,the association between the expression of ATB and clinical characteristics was investigated.Cell proliferation was assessed using a cell counting kit? 8 assay and cell migration was assessed using a wound healing assays.Epithelial? mesenchymal? transition and mitogen? activated protein kinase signaling pathway activity was examined using western blotting.ResultsIt was demonstrated that ATB was highly expressed in lung cancer tissues compared with noncancerous tissues,and associated with tumor size and metastasis.It was also demonstrated that ATB was highly expressed in four lung cancer cell lines compared with the HBE? 1 cell line.The high expression of ATB in A549 and HCC827 lung cancer cell lines was particularly significant.Thus we use them for further research.Cell level experiments showed that the proliferation rate of lung cancer cells using shRNA interference was significantly reduced;while the wound? healing assay showed that the blank band of normal lung cancer cells was narrow,and the blank band of lung cancer cells after shRNA transfection was wider,indicating that shRNA transfection The metastatic invasion ability of lung cancer cells is significantly lower than that of normal cells,which also indicates that the inhibition of lncRNA ATB expression can significantly reduce the metastatic invasion of lung cancer cells.In addition,we also found that inhibition of lncRNA expression significantly increased E-cadherin and decreased N-cadherin expression in both lung cancer cell lines.Conclusions1.The expression level of LncRNA ATB in lung cancer tissues was significantly higher than that in adjacent normal tissues.The expression level of LncRNA inlung cancer cells was significantly higher than that in normal tracheal epithelial cells,indicating that it was highly expressed in lung cancer.2.After silencing with shRNA targeting lncRNA ATB,the proliferation of lung cancer cells was significantly weakened,indicating that silencing its expression significantly inhibited the proliferation of lung cancer cells.3.Silencing lncRNA ATB significantly inhibited the migration and invasion of lung cancer cells.4.Inhibition of lncRNA expression significantly increased E-cadherin and decreased N-cadherin expression in both lung cancer cell lines,thereby confirming that inhibition of lncRNA significantly inhibited p38 expression.Therefore,lncRNA may promote the proliferation and metastasis of lung cancer by regulating E-cadherin and p38 expression.5.High expression of LncRNA ATB suggests a poor prognosis in lung cancer.
