Objective:To evaluate the effects of different hemoglobin,triglyceride,and bilirubin plasma samples on the sensitivity of the latest generation of domestic magnetic bead HBV DNA detection reagents.Methods:HBV-negative blood samples with different clinical plasma traits were selected and HBV national standard products were added according to a certain ratio,so that the final concentration of HBV DNA in the sample reached the minimum detection limit of the detection reagent.The samples were quantitatively tested for hemoglobin,triglyceride and bilirubin,and the samples were grouped according to the detection concentration,and the samples with normal traits were used as controls.Then,HBV DNA nucleic acid detection was performed,calculating the detection rate of HBV DNA in each group,and summarizing the Ct values of the nucleic acid detection(using paired t-test).Results:HBV DNA could not be completely detected when plasma free hemoglobin concentration was 10.13±1.07 g/L.The detection rate of HBV DNA in high triglyceride and hyperbilirubin group was not affected.There was no significant difference in Ct value.(P<0.05).Conclusion:When using the latest generation of domestic magnetic bead HBV DNA detection reagent,lipemia,jaundice and mild to moderate hemolysis samples can be detected normally,but when hemoglobin reaches the clinical severe hemolysis standard,the test results may have false negatives and should be re-draw a blood sample.
