| Objective:Explore the biocompatibility and osteogenesis of sulfonated and graphene oxide modified sulfonated carbon fiber reinforced polyetheretherketone composites;To evaluate the biological safety of the important organs in rats after implantation of experimental materials.Methods:Forty-eight healthy male SD rats were randomly divided into three groups to establish a rat skull defect model.Carbon fibers reinforced polyetheretherketone(CF/PEEK),sulfonated carbon fibers reinforced polyetheretherketone(S-CF/PEEK),graphene oxide modified sulfonated carbon fibers reinforced polyetheretherketone(GO-S-CF/PEEK)materials were implanted according to the experimental grouping,of which CF/PEEK is the control group,S-CF/PEEK,GO-S-CF/PEEK is the experimental group,and the control group and experimental group experimental animals were given fluorescent staining 3 weeks and 1 week before the death,respectively.Twenty-four animals were executed at 8 and 12 weeks after surgery,8 in each group,and Micro-CT was taken to observe the osseointegration in the bone defect areas of each group at different time points.Non-decalcified bone implant sections were made.The fluorescence staining was observed under a laser confocal microscope.After HE staining,the osteogenesis was observed under an opticalmicroscope;after execution,rat heart,liver,spleen,lung,kidney and other internal organs were dissected out,and HE staining was performed to observe the cell morphology of each organ and evaluate the biological safety of experimental materials.The experimental data was analyzed and processed by SPSS 20.0 statistical analysis software.Results:General observation: no animal bleeding,death or other accidents occurred during the whole experiment.Skull specimens were obtained at 8 and 12 weeks after the operation,and the experimental materials were well combined with bone tissue without shedding and bleeding.Micro-CT results showed that new bone formation could be seen on the material surface at 8 and 12 weeks,and the bone binding of the GO-S-CF/PEEK group was better than that of S-CF/PEEK and CF/PEEK group.The fluorescence measurement results showed that the fluorescence markers in each group at 8 weeks were significantly thicker and continuous than those at 12 weeks,with wider fluorescence spacing.At 8 and 12 weeks,mineral apposition rate of GO-S-CF / PEEK group was higher than that of S-CF / PEEK group(P<0.05),and S-CF / PEEK group was higher than CF / PEEK group(P<0.05).Histological observation showed that materials and bone in each group could form micromechanical chimerism,and the surface effect of GO-S-CF/PEEK materials was the best at 12 weeks.HE staining of rat internal organs:there was no significant difference in the morphology of the samples of heart,liver,spleen,lung and kidney in each group,and no abnormal changes such as cell degeneration,hyperemia and edema were observed.Conclusion:Short-term observation of S-CF / PEEK and GO-S-CF / PEEK did not damage important organs in rats;GO-S-CF / PEEK has good biocompatibility and has potential clinical application value. |
