Background Ginkgo biloba L.is the only surviving species in the Ginkgo of the Ginkgoaceae family.It is native to China and is classified as an endangered species.Ginkgo biloba injection raw material is extract of Ginkgo biloba for injection.It is subjected to quality control according to the standard method of Ginkgo biloba extract in Chinese Pharmacopoeia.The chemical composition is diverse and the composition is complex.In particularly,flavonoid glycosides have similar structures and small polar differences,making it difficult to obtain such compounds with clear composition.At the same time,quality control becomes very difficult because it is difficult to obtain a prototype flavonoid glycoside with sufficient purity.Therefore,in order to obtain flavonoid glycosides with sufficient purity,and to further clarify the material basis of extracts and injections.In this paper,the raw materials of Ginkgo biloba injection are selected as the research object,focusing on the separation and purification of flavonoid glycosides.Methods Through a variety of separation and purification means(silica gel column chromatography,gel column chromatography(Sephadex LH-20),reversed-phase mediumpressure column chromatography,semi-preparative HPLC,and preparative LC,etc.)to systematically separate and purify the chemical components.A variety of chromatographic identification techniques(including 1D,2D NMR,ESI-MS,HR-ESI-MS,X-ray crystallographic,IR,UV and CD,etc.)are used to analyze the structure of the isolated compounds.The antiplatelet aggregation rate is determined by the microplate reader method,and the preliminary activity screening of new compounds is carried out.Results Two new coumarinyl flavonoid glycosides are composed of flavonoid aglycones linked to glucose,rhamnose and cis coumarinyl fragments.A novel sesquiterpene lactone is composed of 2,2-dimethylacetone fragment,cyclopentane and lactone ring.The biological activity of anti-platelet aggregation of the isolated new compounds was investigated,and it was found that there was no obvious anti-platelet aggregation activity.Conclusion In this paper,systematic research on the chemical composition and activity of the Ginkgo biloba injection raw material not only improves the chemical composition of the Ginkgo biloba injection raw material,but also improves the utilization efficiency of the Ginkgo biloba leaves extract and preparation.It laid the foundation for further research on Ginkgo biloba preparation and provided a scientific basis for the further clarification of the material basis of Ginkgo biloba injection.
