Objective To investigate the effect of ?-poly-lysine on the formation of Candida albicans persisters,and to explore the possible mechanism of ?-polylysine on the formation of Candida albicans persisters,in order to better control Invasive fungal diseases and drug resistance caused by Candida albicans provide theoretical basis.Methods Collecting and identifying Candida albicans,constructing its mature biofilm,screening Candida albicans persisters for optimal drug concentration,counting the number of colony survival,and verifying the accuracy of persisters by re-testing the minimum inhibitory concentration and prolonging the drug action,besides Candida albicans clinical strains and non-Candida albicans viable counts were counted in colony counting.At the same time,the changes of reactive oxygen species in the control group and the persisters were compared by the fluorescence spectrophotometer,Matrix-Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry was used to determine the changes of the persisters protein profile.Results 55 strains of Clinicall Candida albicans obstetrics accounted for 27.3%,intensive care units accounted for 18.2%,and gynecology accounted for 12.8%,and 55% of the specimens were mainly from sputum specimens,followed by vaginal secretions(29%),9.1% The specimens were derived from fecal specimens.The 55 clinical Candida albicans,40% are strains with weak biofilm formation ability and 34% are medium strains with biofilm formation ability.The optimal concentration of Candida albicans persisters was 102.4mg/mL.At this concentration,the average number of persisters of the Candida albicans(ATCC 64550)was 20CFU/100 uL,the verification of the persisters was confirmed existence(p>0.05).The average number of Candida albicans' s persisters was 90CFU/100 uL,Candida krusei(ATCC 6258)was 10CFU/100 uL,Candida auris was 160CFU/100 uL.At the same time,the content of reactive oxygen species in the persisters group was significantly higher than that in the control group(p <0.05),in addition,the protein peak of the persisters mass spectrum has also clearly changed.Conclusion Obstetrics,gynecology,intensive care units candida albicans have a relatively high clinical isolation composition,and more than 70% of Candida albicans can produce biofilms.There were persisters after ?-PL acts on Candida albicans,and persisters of Candida albicans increases their ability to tolerate reactive oxygen species...
