Objective: The purpose of this study was to analyze the difference of serum human epididymis secretory protein 4(HE4)autoantibody level in patients with lung cancer(LC)and the control group,and to investigate its diagnostic value in high-risk groups of lung cancer.Methods: A total of 61 serum samples from patients with LC,treated in China-Japan Union Hospital of Jilin University from May,2019 to September,2019,were included in this study,and another 53 serum samples from sex,age,and smoking history matched 29 healthy donors and 24 benign lung diseases(BLD)patients were collected as the control group,among which all 61 patients with LC had specific pathological results,including 44 ADC,15 SCC and 2 SCLC.Analyzed them by Enzyme-linked Immunosorbent Assay,ELISA),established the receiver operating characteristic(ROC)curve to estimate the diagnostic value of HE4 antibody,and analyzed the difference of serum HE4 autoantibodies level in LC groups and control group.According to the result of pathology,analyzed HE4 autoantibodies level in disparate pathological subtypes and clinical stages.Results: According to detection,the ROC curve demonstrated a 67.21% sensitivity and a 96.23% specificity along with a AUC of 0.848(95% CI:0.776-0.920)for HE4 autoantibody,and for the diagnosis of early lung cancer,the sensitivity was 54.76%.Meanwhile,there were significant differences of diagnostic value between patients with LC and control group(0.487±0.248 vs 0.208±0.098,Z=6.389,p<0.001)and between healthy donors and patients with benign lung diseases patient(0.247±0.124 vs 0.175±0.056,Z=-2.314,p=0.021).Among different pathological subtypes,the level of HE4 autoantibody was higher in adenocarcinoma than in squamous cell carcinoma(0.535±0.260 vs 0.336±0.154,Z=-2.768,p=0.006),and serum HE4 autoantibody in the patients with III-IV stages lung cancer was higher than the patients with I-II stages lung cancer(Z=2.297,p=0.022).Conclusion: Serum HE4 autoantibody is an important biomarker for the diagnosis of LC,and the detection of serum HE4 autoantibody might have potential diagnostic value in high-risk groups of LC.It provides a new method for the diagnosis of lung cancer in the clinic.
