| Objective: to screen out the differentially expressed miRNAs in the intestinal type gastric carcinoma group and the diffuse type gastric carcinoma group,and to analyze and select the miRNAs with large difference for further study.Then,the selected miR-1246 was discussed to analyze whether the expression levels of miR-1246 and dennd2 d,one of the predicted target genes,were different between intestinal type gastric carcinoma and diffuse type gastric carcinoma,the expression levels of miR-1246 and dennd2 d and their clinical correlation were analyzed,and the correlation was analyzed.Methods:(1)a total of 112 gastric carcinoma tissue specimens and corresponding gastric carcinoma paracancer tissue specimens were collected,including 58 intestinal type gastric carcinoma specimens and 54 diffuse gastric carcinoma specimens.Six cases of intestinal type gastric cancer,diffuse type(non-mucinous type)and diffuse type(mucinous type)gastric cancer were selected.miRNAs was extracted from the 18 samples,and miRNAs microarray gene chip analysis was performed to obtain miRNAs expression profiles in gastric cancer tissues.miRNAs with large differences were analyzed,literatures were consulted and the research value of these screened miRNAs were analyzed,and miRNAs that needed further study were selected according to their scientific research value.(2)from 112 gastric carcinoma specimens of paraffin tissue extract miRNAs,reverse transcription,adopts the method of RT-PCR detection of intestinal type of gastric carcinoma and diffuse gastric carcinoma group in the relative expression level of miR-1246,and analyses its relationship with clinical pathological features.(3)112 samples were microchip and immunohistochemical staining was used to compare the expression levels of dennd2 d in the intestinal type gastric carcinoma group and the diffuse type gastric carcinoma group,and the relationship between the clinical and pathological characteristics was analyzed.The relevant clinical data,the literatures related to miR-1246 and denn2 d were consulted,and the statistical analysis was conducted in combination with the detection results.Results :(1)A total of 22 differentially expressed miRNAs were screened between the intestinal type gastric cancer group and the diffuse(non-mucinous)gastric cancer group,of which 19 miRNAs were up-regulated in the intestinal type gastric cancer tissue,while the remaining 3 were up-regulated in the diffuse gastric cancer tissue.A total of 60 differentially expressed miRNAs were screened between the intestinal type gastric cancer group and the diffuse(mucinous)gastric cancer group,of which 21 miRNAs were up-regulated in the intestinal type gastric cancer tissue,while the remaining 39 were up-regulated in the diffuse gastric cancer tissue..(2)The RT-PCR detected that the expression of mir-1246 in the intestinal type gastric cancer group was higher than that in the diffuse type gastric cancer group(P <0.05).The expression of miR-1246 in the enterogastric carcinoma group was statistically significant with tumor grade,TNM stage and lymph node metastasis(P<0.05),and the expression in the diffuse gastric carcinoma group was statistically significant with tumor TNM stage(P<0.05).(3)there was no difference in the expression of dennd2 d in the intestinal type gastric carcinoma group and the diffuse type gastric carcinoma group.The expression of dennd2 d in the intestinal type gastric carcinoma group was statistically significant with ethnic and lymph node metastasis(P<0.05),and the expression in the diffuse type gastric carcinoma group was statistically significant with the location of tumor occurrence(P<0.05).Conclusion :(1)there were significant differences in the expression of miRNAs between the intestinal type gastric carcinoma group and the diffuse type gastric carcinoma group.(2)miR-1246 is highly expressed in human intestinal type gastric cancer and low in diffuse type gastric cancer.The determination of the level of mir-1246 in the tissues of patients with gastric cancer can provide a direction for the diagnosis and treatment of tissue types.(3)The expression of mir-1246 in intestinal gastric cancer tissues is correlated with the presence or absence of metastasis of lymph node cancer and TNM stage,so it can be speculated that mir-1246 is associated with lymph node metastasis,which can provide ideas for the evaluation of lymph node metastasis status and the determination of the stage and progress of gastric cancer.(4)The expression of dennd2 d in intestinal type gastric cancer is correlated with tumor size,tumor TNM stage and lymph node metastasis.It is speculated that dennd2 d may provide some basis for judging the progress of gastric cancer and the stage of tumor. |
