Objective: 1.To investigate the role of thrombospondin-1(TSP-1)and hedgehog signaling pathway(Hh)in mucoepidermoid carcinoma of salivary gland;2.To explore the potential effect of TSP-1 on the key genes Sufu and GLI1 of the Hedgehog signaling pathway in mucoepidermoid carcinoma.Methods: 1.From 2010 to 2019,52 cases of mucoepidermoid carcinoma and 10 cases of normal salivary gland were divided into experimental group(52 cases of mucoepidermoid carcinoma)and control group(10 cases of normal salivary gland).The pathological results were reexamined by hematoxylin and eosin(HE)staining;the expression of SuFu,GLI1 and TSP-1 protein in mucoepidermoid carcinoma and normal salivary gland were detected by immunohistochemical staining,and the relationship between the three proteins and the clinical,imaging and pathological characteristics of mucoepidermoid carcinoma was analyzed;2.The mucoepidermoid carcinoma MC-3 cell line(oral cavity of Air Force Military Medical University)was selected Biology teaching and research office established and provided)as the research object,the cells were divided into blank control group,TSP-1 action group,TSP-1+CD36 blocking group,and the effects of TSP-1 on the protein expression of the key genes Sufu and GLI1 in the process of differentiation and proliferation of mucoepidermoid carcinoma cells were studied by immunofluorescence detection and fluorescence densitometry(Image J);3.Chi square test and Fisher exact probability method were used for the counting data;and mean ± standard deviation was used for the measurement data;multiple sample mean single factor ANOVA was used for the comparison between multiple groups,and independent sample t-test was used for the comparison between the two groups;P < 0.05 was statistically significant.Results: 1.In MEC group,the positive rates of TSP-1,Sufu and GLI1 were 63.5%(33/52),51.9%(27/52),57.7%(30/52),the positive expression rate of TSP-1,Sufu and GLI1 protein in normal salivary gland group was 10%(1/10),the difference between the experimental group and the control group was statistically significant(P<0.05);2.thepositive expression rate of TSP-1 in high,medium and low differentiated MEC decreased in turn,the difference between high and low differentiated MEC was statistically significant(P<0.05);The positive expression rate of Sufu in high,medium and low differentiated MEC increased in turn,and the difference of protein expression rate in high and low differentiated MEC was statistically significant(P<0.05);At the same time,the positive expression rate of GLI1 in high,medium and low differentiated MEC was also significantly higher,and the difference between high and low differentiated MEC was statistically significant(P<0.05);3.The expression level of TSP-1 protein was closely related to the difference of MEC tumor CT(Computed Tomography,CT)diameter and neck lymph node metastasis(P<0.05),but not to smoking,drinking,age,location,clinical stage,clinical diameter,primary and recurrence and other clinical characteristics(P>0.05);the expression level of Sufu protein was closely related to MEC tumor CT diameter and neck lymph node metastasis(P<0.05);but not to smoking,drinking,age,location,clinical stage,clinical diameter,primary and recurrence and other clinical characteristics(P>0.05);The expression level of GLI1 was closely related to the difference of MEC tumor CT diameter and cervical lymph node metastasis(P<0.05),but not to the factors of smoking,drinking,age,position,clinical stage,clinical diameter and primary and recurrence(P>0.05);4.In the blank control group,the average optical density value of GLI1 and Sufu protein expression was the highest,the average optical density value of TSP-1 protein group was the lowest,the average optical density value of TSP-1+CD36 blocking group was between the two,the difference of the average optical density value of Sufu and GLI1 protein between the three groups was statistically significant(P<0.05);the difference of protein expression between TSP-1 group and blank control group was statistically significant(P<0.05);The difference of protein expression between TSP-1 group and TSP-1 + CD36 blocking group was statistically significant(P<0.05).Conclusion: In mucoepidermoid carcinoma,TSP-1 protein may affect the abnormal activation intensity of hedgehog signaling pathway by inhibiting the protein expression of Sufu and GLI1 through receptor CD36,thus inhibiting the occurrence and development of mucoepidermoid carcinoma.However,by binding with CD36 receptor,TSP-1 protein may affect the expression activity of SHH,the upstream hedgehog signaling pathway promoter of Sufu and GLI1,which may inhibit mucoepidermoid carcinoma The key point is,but the specific mechanism needs further study.
