Clinical Application Of Mycobacterium Tuberculosis Rv1860,Rv0494 And Rv1876 Proteins In The Diagnosis Of Active Tuberculosis

Purpose:The clinical application value of candidate biomarker Rv1860,Rv0494,Rv1876 proteins in active tuberculosis diagnosis is evaluated by protein chip technology,providing a new idea for the diagnosis of active tuberculosis.Methods:120 serum samples are collected from active tuberculosis patients?ATBI group?,latent tuberculosis patients?LTBI group?,disease control patients?D group?and healthy volunteers?N group?who were admitted to the Affiliated Hospital of Zunyi Medical University from January 2018 to March 2019.Each group contains 30 serum samples.The protein microchips customized by Rv1860,Rv0494 and Rv1876 proteins at different concentrations are used for antigen-antibody reaction with 120 serum samples.The effective data are normalized,and the AUC of Rv1860,Rv0494 and Rv1876 proteins are analyzed and calculated based on Rv1860,Rv0494 and Rv1876 proteins alone,combined with each other and combined diagnosis of the three proteins,so as to evaluate their clinical application value in the diagnosis of active tuberculosis.Results:?1?Rv1860,Rv0494,Rv1876 are used in distinguishing active tuberculosis from latent tuberculosis infection:for the serum IgG immune response,the AUC of Rv1860_y25 is 68.2%;the AUC of Rv0494_y is 67.7%;the AUC of Rv1876_y is57.2 %;the AUC of Rv1860 combined with Rv0494 is 73.0%;when the three indicators are diagnosed in combination,AUC is 76.8%;?2?Rv1860,Rv0494,Rv1876 are used to identify active tuberculosis and pneumonia,lung cancer:for the serum IgG immune response,the AUC of Rv1860_y25 is 70.1%;the AUC of Rv0494_y is 69.8%;the AUC of Rv1876_y is 61.7 %;the AUC of Rv1860 combined with Rv0494 is 73.6%;when the three indicators are diagnosed in combination,AUC is74.4%;?3?Rv1860,Rv0494,Rv1876 are used to identify active tuberculosis and healthy people:for the serum IgG immune response,the AUC of Rv1860_y25 is 75.6%;the AUC ofRv0494_y is 77.3%;the AUC of Rv1876_y is 62.9%;the AUC of Rv1860 combined with Rv0494 is 79.0%;when the three indicators are diagnosed in combination,AUC is 80.0%.Conclusion:?1?For the serum IgG immune response,the combination of Rv1860,Rv0494 and Rv1876 can be used to distinguish between active tuberculosis and latent tuberculosis infection.?2?For the serum IgG immune response,the combination of Rv1860,Rv0494 and Rv1876 can be used to distinguish active tuberculosis from pneumonia,lung cancer.?3?For the serum IgG immune response,the combination of Rv1860,Rv0494 and Rv1876 can be used to distinguish active tuberculosis from healthy people,which could be used in the auxiliary diagnosis of active tuberculosis.
