In recent years,the rapid development of 3D bioprinting technology has become an important research field in tissue engineering and biomedicine.As a typical soft material,bioink is directly related to the final molding effect,the desired accuracy and biocompatibility of printing.The decellularized matrix(DECM),derived from natural tissues,has the main biological characteristics of the original liver tissue,so it has good biocompatibility,degradability,and ability to induce angiogenesis and tissue regeneration,becoming the preferred material for bioink.Current research has found that the printability of the decellularized matrix is not good,and can only be used as an additive to bioink,which results in the limitation of suitable natural growth factors and active biological components in the printed construct,affecting the activity of normal cells.Based on the above background,this paper studies a new type of bioprinting composite material,whose main components include nanoclay,rat liver decellularized matrix and methacrylic anhydride gelatin(GelMA).Among them,the liver decellularized matrix is obtained from rat liver tissue through the joint perfusion of Triton X-100 and sodium laurylsulfate(SDS),which is rich in collagen,glycosaminoglycans,growth factors bFGF,HGF and many other important ingredients.The experimental results show that,due to the addition of nanoclay,not only the storage modulus of the bioink has been improved,it has excellent rheological properties,and the printed 3D scaffold has a clear structure,which has good printing performance,and it even increased the content of decellularized matrix of rat liver to 75%,which is the highest content reported in the literature.It was found that the primary hepatocytes growing on the surface of the 3D cell scaffold printed with this composite material had good growth activity and albumin expression.Specificall,,the albumin secretion content of primary hepatocytes is 9.35g/L on the first day of culture,which is significantly higher than the secretion content of other components of bioink(P <0.01),indicating that the bioink has a good biocompatibility,can effectively improve the survival and proliferation of primary hepatocytes,and has low cytotoxicity.
