| Objective:From the perspective of abnormal copy number,this paper discusses the role of cell cycle related genes in the occurrence and development of high-grade serous ovarian cancer,to find out the target genes that affect the prognosis of the disease,and to provide a reference for drug selection in clinical treatment in the future.Methods:1.Differential expression genes of high-grade serous ovarian cancer were obtained from GEO database.2.Copy number variation genes of high-grade serous ovarian cancer were obtained from TCGA database.3.The pathways and genes related to cell cycle were screened from GO,KEGG,REACTOME and BIOCARTA databases.4.To integrate genes related to cell cycle,copy number variation and differential expression in high-grade serous ovarian cancer.5.The target genes that affect the occurrence and development of the disease were obtained by combining the survival curve.Results:1.827 up-regulated genes and 950 down-regulated genes were screened from GEO database with p-value of 10-9 as the cut-off value.2.A total of 1999 copy number amplification(AMP)genes and 47 copy number deletion(DEL)genes were screened from TCGA database with 10%differential expression frequency as the cut-off value.3.178 cell cycle related pathways and 3213 cell cycle related genes were identified from GO,KEGG,REACTOME and BIOCARTA databases.4.36 genes related to the cell cycle of high-grade serous ovarian cancer were screened out.5.Combined with the analysis of survival curve,six genes including ECT2,RAD51AP1,YWHAZ,RAD21,SMC4 and TFB2M were found to be significantly related to the poor survival prognosis of high-grade serous ovarian cancer.Conclusion:1.The occurrence of some high-grade serous ovarian cancer may be caused by the up-regulation of cell cycle related genes through the amplification of copy number,thus affecting cell cycle.2.ECT2,RAD51AP1,YWHAZ,RAD21,SMC4 and TFB2M are genes related to cell cycle with copy number variation and differential expression,and are significantly related to poor survival prognosis of high-grade serous ovarian cancer.3.MECOM,PRKCI and ECT2 are genes related to cell cycle with copy number variation and differential expression,and may share the same signal pathway to promote the occurrence and development of high-grade serous ovarian cancer. |
