Objective:To investigate the effect of rapamycin on proliferation and apoptosis of acute monocytic leukemia SHI-1 cells in vitro and in vivo.Methods1.Human acute monocytic leukemia cell lines SHI-1 cells were divided into two groups:negative control group and rapamycin group(the final concentration was5uM,10uM,20um,respectively).SHI-1 cells were treated with different concentrations of rapamycin.The inhibitory effect of rapamycin on the proliferation of SHI-1 cells was detected by CCK8 assay at 24 h,48 h and 72 h.After 24 hours of drug treatment,apoptosis was detected by flow cytometry.2.SHI-1 cells were subcutaneously inoculated to the nude mice to establish animal model.The nude mice were subcutaneously inoculated with 5×10~6 SHI-1 cells and randomly divided into two groups:control group(n=6)and treatment group(n=6).Rapamycin(10mg/kg)was injected into the tumor 10 days after subcutaneous tumors were formed,and the tumor size was measured every five days.All nude mice were killed 25 days after inoculation.Results:1.Rapamycin could significantly inhibit the proliferation of SHI-1,and the proliferation inhibition rate increased in a dose-and time-dependent manner.Compared with the negative control group,SHI-1 cells treated with rapamycin of5uM for 24 hours could not induce apoptosis.When the concentration of rapamycin increased to 10uM,it could significantly induce apoptosis,and the proportion of apoptosis in 20uM rapamycin group increased significantly.2.The subcutaneous suspension of SHI-1 cells was completely absorbed 2 days after inoculation.On the 5th day,subcutaneous tumor formation could be seen by eyes.The subcutaneous tumors of nude mice in the control group grew logarithmically.Compared with the control group,the growth of subcutaneous tumor of the nude mice in the treatment group was significantly slowed down,and the tumor volume was significantly smaller than that of the control group.On the first,fifth and tenth days after treatment,the average subcutaneous tumor volume of the two groups was 483.52±198.58mm~3,915.90±343.56mm~3,1371.75±406.72 mm~3(control group),273.08±114.111mm~3,583.49±188.90mm~3,1015.90±249.77mm~3(treatment group),and the tumor inhibition rate was about 43.52%,36.29%and 25.94%respectively.Conclusions1.Rapamycin could significantly inhibit the proliferation and induce apoptosis of SHI-1 cells in a time-and dose-dependent manner.2.Rapamycin could effectively inhibited the proliferation and growth of SHI-1cells in nude mice.
