Objective:This article intends to introduce a hyaluronic acid-gold optical probe and discuss the research mechanism of dual-mode detection of Osteoarthritis.Method:1.One-step synthesis of “Hyaluronic acid-gold nanoprobe(HA-GNP),Thiolated hyaluronic acid(HS)and rhodamine B(RHODAMINE B)fluorescent dye selfassembled synthesis of “hyaluronic acid-gold Nano-RHODAMINE B fluorescent probe "(HA-HS-GNP-RHODAMINE B);2.The Observation of GNP,HA-GNP,HA-HS-GNP-RHODAMINE B was indentified by UV spectrophotometer,DLS,Zeta potential,scanning,transmission electron microscope observation;3.Building two detection methods based on ROS colorimetric method and fluorescence method in detecting OA disease,with exploring its mechanism of action;4.Constructing an in a vitro cell model,the experiments determine the cytotoxicity of the probe using CCK-8 and fluorescent staining of living dead cells;5.Detect the potential therapeutic ability of the probe by constructing in vivo animal time.6.Identifying the traceability of HA-HS-GNP-FITC to OA inflammatory cells under a fluorescent microscope.Results:1.Hyaluronic acid-gold nanoparticles(HA-GNP)are more stable than traditional sodium citrate-gold nanoparticles(GNP);2.The stability difference and color change of HA-GNP and GNP can be used as the basis for ROS related colorimetric detection;3.Based on the fluorescence change of HA-HS-GNP-RHODAMINE B probe under ROS,reveal the role of fluorescence detection4.HA-GNP / HA-HS-GNP-RHODAMINE B has good biocompatibility and is suitable for in vivo and in vitro experiments and clinical applications5.The tracing effect of HA-HS-GNP-FITC on OA chondrocytes may be used as a new method for clinical OA cell detection.Conclusion:1.Hyaluronic acid-gold nanoparticles(HA-GNP)are more stable than traditional sodium citrate-gold nanoparticles(GNP);2.Successfully constructed hyaluronic acid-gold nanoprobe(HA-GNP)and thiolated hyaluronic acid-gold-RHODAMINE B fluorescent probe(HA-HS-GNPRHODAMINE B);3.The probe is more stable than traditional gold nanoparticles;4.Explore the detection methods of the two probes based on the colorimetric and fluorescent probes under ROS;5.The two probes have low cytotoxicity and have certain tracing ability to OA cells;and it has a potential therapeutic effect on chondrocytes;...
