Objective: To explore the role of TRAF6 in the reduction of HNE-induced mucin MUC5AC expression in CRSwNP caused by MiR-146 a.Methods: 1?IHC,qRT-PCR,ELISA to detect the secretion of MUC5AC,MiR-146 a and TRAF6 in the nasal cavity and sinus mucosa of CRSwNP group and normal control group;2?Primary epithelial cells of nasal mucosa were cultured in vitro,and after stimulation with HNE,the protein and m RNA secretion of MUC5AC,MiR-146 a,TRAF6 were detected by WB,ELISA,qRT-PCR,TRAF6 were detected by WB,ELISA,qRTPCR;After transfection of nasal mucosal epithelial cells stimulated with HNE with MiR-146 a NC,MiR-146 a mimic and MiR-146 a inhibitor,WB,ELISA,qRT-PCR were used to detect the protein and m RNA secretion of MUC5AC,MiR-146 a,TRAF6;3?After TRAF6 inhibitor acted on MiR-146 a inhibitor transfection group,the expression trend of MUC5AC and TRAF6 was opposite to that of mi R-146 a inhibitor group;4?Identification of the relationship between TRAF6 and MiR-146 a by dual luciferase reporter gene.Results: 1?Compared with controls,TRAF6 and MUC5AC in CRSwNP were up-regulated by IHC,qRT-PCR and ELISA(P <0.05);MiR-146 a was down-regulated;2?In the HNE(10n M,24h)group,MiR-146 a,TRAF6,and MUC5AC were upregulated compared to the control group;in the MiR-146 a mimic group,MiR-146 a was significantly increased over the NC group,and TRAF6 and MUC5AC were down-regulated compared to the NC group;MiR-146 a inhibitor group In the opposite trend;3?After TRAF6 inhibitor action,the secretion trend of TRAF6 and MUC5AC showed the opposite trend to the MiR-146 a inhibitor group(p <0.05);4?The RLUC value measured by the mi RNA mimics group was significantly lower than that of the internal control group;the MiR-146 a inhibitor group had the opposite result,indicating that TRAF6 is the target gene of MiR-146 a.Conclusions: MiR-146 a inhibits HNE-induced MUC5AC overexpression by targeting and regulating TRAF6 secretion.
