Objective:Aldrichina grahami was considered as an object of study to explore the effects of methamphetamine(MA)on the development of A.grahami.This study provides a theory for forensic scientists to estimate the postmortem interval(PMI)and the cause of death by using the development data of A.grahami in drug-related cases. Methods:The adults of A.grahami were trapped in a wild and established the purebred colony in a laboratory. The developmental data of A.grahami were collected and analyzed on rabbit mince at four different concentrations of MA(0,45,90,180 ng/mg).The gaschromatography-mass spectrometry(GC-MS)was established to detect the MA content of A.grahami at different stages.Results:(1)in larval instar, the average length of A.grahami in MA groups was longer than that in the control;the T3 group was retarded about 10 h compared with the C;during the pupation period(P),the T3 had lower body weights about 6.66 mg than those of C,and the weights of adults in T3 were heaveir 4.36 mg than those of C.MA had effect on the survival rate of pupation, which was statistically significant.(2)The evaluation of standardized methods:the contents of MA were showed a good linear relationship with the peak area in the range of 0.10-40.00ng/mg,R~2>0.9998,the LOD was 0.10 ng/m L,the LOQ was 0.33 ng/m L,and the CV was7.70-19.20%.(3)The concentrations of drug in the flies at different stages:MA showed the higher the concentration in rabbit mince along with the higher the cumulative concentration in flies. The concentrations of MA revealed an upward tendency before the larvae reached the third instar(L3).To note, the concentrations of MA were successfully detected from empty puparium(EP),and the concentration of EP was close to pupae. Conclusion:MA has an effect on the development of A.grahami.The changes of MA content in flies were related to their physiological metabolism.
