Objective: Graphene oxide(GO)/Egg white(EW)/Polyvinyl alcohol(PVA)fibrous wound dressings with different GO concentrations were fabricated by electrospinning method,and study its application potential in skin tissue repair to offers a new option for full-thickness skin defect treatment in clinic.Methods: The conductivity and viscosity of four GO/EW/PVA mixed solutions with GO content of 0,0.5,1,2 mg/ml were measured,and the general morphology of the four dressings and the fiber morphology in the scanning electron microscope and transmission electron microscope were observed,to evaluate spinnability of the four groups of mixed solutions.The prepared fibrous dressing was measured for its hydrophilicity,mechanical properties,cell compatibility,and healing effect of skin full-layer defects in vivo.Results: As the GO content increased,the viscosity of the electrospinning solution increased and the conductivity decreased.Excessive viscosity increases the number of microsphere fibers of the dressing,and causes the surface of the fiber dressing to be rough and a large number of droplets deposited,making the preparation of the fiber dressing difficult.TEM showed that GO successfully adhered to the fiber.FTIR and XRD confirmed that GO and EW were successfully combined with PVA fibers,and the crystallinity of ovalbumin increased slightly after cross-linking.When the GO content is 1 mg/ml,the wound dressing has the best comprehensive performance,not only has good hydrophilicity,thermal stability,tensile mechanical properties,but also good cell compatibility,which can increase cell survival rate and promote proliferation,adhesion,migration and morphological changes of human skin fibroblasts(HDFs)and human umbilical vein endothelial cells(HUVECs).In vivo healing experiments further showed that a wound dressing with a GO content of 1 mg/ml had the best promotion effect on healing of full-thickness skin defects and skin tissue regeneration.Compared with the other groups,after 10 days,the wound closure in this group was the most significant.At 14 days,the neonatal epidermal tissue was smooth and there were no red,uneven scars on the wound.On the 7th day of HE staining,there were more capillaries and extracellular matrix in this group,and the skin regeneration was enhanced.On the 10 th day,reconstructed skin tissue with neonatal accessory glands(such as hair follicles)appeared;on the 14 th day,the skin accessory glands(Hair follicles)and structures closer to normal skin tissue.Compared with the other groups,Masson staining on the 14 th day showed that GO had a densest collagen deposition in the wound area of 1 mg/ml group,and CD31 staining on the 7th day showed a significant increase in neovascularization density.Conclusion: GO/EW/PVA electrospun fibers with GO concentration of 1 mg/ml have great potential in wound dressing applications.As a multifunctional wound dressing,they can provide a new option for the treatment of intractable full-thickness skin defects in clinical practice.
