Objectives:In this study,the methods of data mining and network pharmacology were used to study the prescription law of traditional Chinese medicine in the treatment of Ulcerative Colitis(UC)by director Zhou zhenghua,and summarize his clinical experience and academic thoughts on the treatment of UC.The potential target and molecular mechanism of director zhou zhenghua's treatment of UC in combination of basic drugs were preliminarily discussed to provide reference for further experimental research.Research methods:Through the application of TCM inheritance assistant platform software(V2.5)and Microsoft Excel(2016),the medical records of UC patients receiving TCM treatment from September 2017 to December 2019 in the outpatient department of the first affiliated hospital of tianjin university of traditional Chinese medicine,the gastroenterology outpatient department and the ward after being treated by director zhou zhenghua were sorted out and collected.Screening was conducted in strict accordance with the inclusion criteria and exclusion criteria,data of eligible patients were input into the TCM inheritance auxiliary platform system for analysis and processing,prescription medication law of director zhou zhenghua for treating UC was obtained,and his medication experience and clinical experience were summarized.Under the guidance of zhou's teacher,the basic drug combinations in the above studies were selected,and the network pharmacology method was adopted to collect and screen the active ingredients and targets of traditional Chinese medicine,compare them with UC related targets,and screen the common genes by means of Venn diagram.Cytoscape 3.7.2 software was used to make the "active component-target-disease network map of basic drugs" based on the above data.Based on the PPI database of the protein interaction network,the target interaction network was constructed to extract the candidate target of the basic drug combination therapy for UC,and the GO enrichment analysis and KEGG pathway enrichment analysis were carried out to preliminarily explore the molecular mechanism of the basic drug combination therapy for patients who met the inclusion criteria,with a total of 309 prescriptions.Data were input into the TCM inheritance auxiliary platform system,and the following results were obtained:the maximum age of UC patients included was 76 years old,the minimum age was 17 years old,and the average age was about 47.6 years old.Among them,58 males and 45 females showed no significant gender difference.Syndrome and activity analysis:the main syndromes of the patients were dampness-accumulation syndrome of spleen deficiency,followed by dampness-heat syndrome of large intestine,spleen-kidney deficiency syndrome of Yang deficiency of spleen and kidney,mixed syndrome of cold and heat,syndrome of heat toxicity,syndrome of liver-stagnation and spleen-deficiency,syndrome of Yin and blood deficiency,mainly in remission period.Analysis of pharmacokinesis of drug taste:director zhou used both cold and warm to treat this disease,and used 1621 times(30.25%)of warm drugs and 1556 times(29.04%)of cold drugs.The main flavor was bitter(32.40%),gan(31.10%)and xin(21.63%).The drug to spleen,stomach meridian is more common Among the 309 prescriptions,there were 97 traditional Chinese medicines.The top 10 herbs with the highest frequency of use were atractylodes,radix paeoniae,cocos,licorice root,coix seed,codonopsis codonopsis,bran Fried coix seed,ground elm,rhizoma coptidis and ginger charcoal.Based on the analysis of the medication law of association rules,it is concluded that the basic medication combination of director zhou zhenghua in the treatment of this disease is codonopsis,atractylodis,pachycocos,licorice root,white peony root,coix seed,bran Fried coix seed,rhizoma coptidis,radix rubediae,and ginger charcoal.Analysis of drug correlation degree and bitter,sweet and astringent taste.The frequently used Chinese medicines were rhizoma coptidis,rhizoma atractylodis,coix seed,white peony root,ground elm,radix puerariae root,beaten sauce,scutellaria,bran Fried coix seed and angelica.The spleen-kidney Yang deficiency syndrome is mainly used in terms of temperature,with a taste of sweet and bitter.The top 10 drugs with high frequency of use were atractylodes,poria cocos,licorice root,radix codonopsis,dried ginger,white peony root,psoraleae,nutmeg and coix seed.2 Research on the molecular mechanism of network pharmacology of zhou zhenghua's combination of basic drugs in the treatment of ulcerative colitisA total of 9 traditional Chinese medicines contained 81 chemical components.All the potential targets of the above components were collected,and 200 potential targets were obtained after sorting out,comparing and deweighting.After searching GeneCards and Digsee databases,a total of 2408 UC related genes were obtained.Compared with the aforementioned targets,137 common genes were obtained.The composition-target relationship was used to construct the basic drug-active component-target-disease network,and 48 active components were obtained.According to the protein interaction network of common targets,AKT1,CASP8,CDKN1 A,IL10 and other targets were found to be in the core position.GO further enrichment of functional analysis,KEGG pathway enrichment analysis shows that the drug combination could by participating in cytokines receptors,receptor ligand activity and cytokine activity,ubiquitin protein ligase combination,RNA polymerase ?transcription factor binding and other biological processes and PI3K Akt,IL-17,toll-like receptors signaling pathways play a role in the treatment of UCConclusions:1 cytokine activity.This study revealed the complex mechanism of treating UC by tutor's combination of basic drugs with UC through multi-target,multi-pathway and multi-pathway,and provided reference for subsequent studies.
