ObjectAt present,the role of autophagy in cell invasion(especially tumor cells)and pregnancy has been reported,and LC3,Beclin1 and p62 are commonly used markers in autophagy research.The occurrence of placenta previa with placenta accreta is closely related to the abnormal invasion of trophoblasts.The purpose of this study was to explore the expression level and correlation of placenta trophoblast autophagy in placenta previa with placenta accreta by detecting the expression level of the above three proteins and genes in placenta previa with placenta accreta.MethodsThe placental tissues of 20 pregnant women with placenta previa and placenta accreta and 20 normal pregnant women who underwent regular antenatal examination in our hospital from April 2018 to January 2020 were collected.In this study,the expression of autophagy-associated proteins LC3,Beclin1 and P62 was detected by immunohistochemistry,real-time quantitative polymerase chain reaction(qRT-PCR)and Western blotting,and the formation of autophagosomes in placental trophoblasts was observed under transmission electron microscope,so as to explore the relationship between autophagy of placental trophoblasts and placenta previa with placenta accreta.Results1.General condition: there was no significant difference in age,pregnancy,delivery,induced abortion and cesarean sect ion between the two groups(P > 0.05).2.Immunohistochemi stry: the average optical density(AOD)of LC3,Beclin1 and p62 in the group of pregnant women with placenta previa and placenta accreta was 0.028±0.019,0.030±0.025 and 0.0033±0.0042,while the AOD of the above three proteins in normal group was 0.012±0.007,0.011±0.006 and 0.0131±0.0112.There were significant differences i n the expression of these three prot eins between the two groups(P<0.05).3.qRT-PCR: the expression of LC3 mRNA and Beclin1 mRNA in the group of pregnant women with placenta previ a and placent a accreta were 4.65 7±3.784 and 2.183±1.668,while those in normal group were 1.800±0.993 and 1.832±1.810.There was significant difference in t he expression of LC3 mRNA between the t wo groups(P<0.05),but no significant difference in the expre ssion of Beclin1 mRNA between the two groups(P>0.05).4.Western Blot: the expression of LC3-II,Beclin1 and p62 protein in the group of pregnant women with placenta previa and placent a accret a was 1.444±0.224,0.750 ± 0.418 and 0.797±0.210,while that in normal group was 0.838±0.046,0.174±0.061 and 1.078±0.153.There were significant differences in the expression of LC3-II,Beclin1 and P62 between the two groups(P<0.05).5.Transmission electron microscope: the proportion of autophagic vacuoles in the cytoplasmic area in the group of pregnant women with placenta previa and placenta accreta was 0.033 ±0.003,while that in normal group was 0.009 ±0.010.There was significant difference between the two groups(P < 0.05).ConclusionIn the trophoblast cells of patients with placenta previa and placenta accreta,the expression of autophagy-related proteins LC3 and Beclin1 were up-regulated,while the expression of p62 was decreased,and the formation of autophagosome increased,indicating that the autophagy activity was enhanced.It suggests that the occurrence of placenta previa with placenta accreta is related to the enhancement of autophagy activity.
