[Objective] To explore the feasibility of near-infrared two-window fluorescent photosensitizer in diagnosis and treatment of oral squamous cell carcinoma.[Methods] NIR-II small molecular NDI-NA was prepared,the structure of NDI-NA was identified by MS and NMR,organic nanoparticles NDI-NA NPs were synthesized by nanoprecipitation method.Morphology and size of NPs were observed by TEM,and particle size were detected by DLS.Absorption and emission spectra of NPs were detected.The stability of NPs was detected under continuous laser irradiation,and study of its photothermal / photodynamic performance was carried out.The study of the tumor suppressive effect of NDI-NA NPs at the cellular level and in animal models were carried out.The biocompatibility of NDI-NA NPs was verified by hemolysis test,analysis of nude mice blood routine and biochemistry and organ structure changes.[Results] Organic nanoparticles NDI-NA NPs were successfully synthesized and prepared.The shape was spherical and the average size was about 27 nm.The maximum absorption peak was 904 nm and the maximum emission peak was 1020 nm.The NPs had excellent photothermal conversion efficiency(~ 30.8%)and Singlet oxygen quantum yield(0.16%).Both in vivo and in vitro experiments confirmed that NDI-NA NPs have excellent tumor suppressing effect.four hours after the tail vein injection of nanoparticles,the NIR-II fluorescence signal was significantly enriched in the tumor.After a single administration,under the irradiation of 808 nm laser,the tumor completely disappeared within 14 days and there was no tumor recurrence.There was no obvious damage to important organs in nude mice,and no significant change in analysis of blood routine and biochemistry.[Conclusion] The organic nanoparticles NDI-NA NPs were successfully prepared,with excellent photothermal conversion efficiency and Singlet oxygen quantum yield(0.16%),stable performance and good biocompatibility.It may be clinically applied on diagnosis and treatment of OSCC in the future as effective means.
