Objective:Analysis of " Three-acupoint and Five-needle Method " Acupuncture Treatment of Whole Blood miRNA and Whole Genome m RNA Expression Profile in Asthmatic Patients,integrate analysis of the role of miRNAs and their target genes in acupuncture anti-asthma process,screening of miRNAs and their target genes for acupuncture anti-asthma,to explore the mechanism of acupuncture against asthma from the perspective of miRNA.Method:According to the Asthma Control Test(ACT)and the patient's medication,10 cases of " Three-acupoint and Five-needle Method " acupuncture treatment were used to study the peripheral blood of asthma patients.The total RNA in peripheral blood was extracted and used separately.Agilent Human miRNA v21.0 chip and SBC Human((4×180K))ce RNA chip were used to detect the expression profiles of miRNAs and m RNAs before and after acupuncture treatment.The target genes were predicted by differential miRNAs using mi RWalk,and m RNA expression profiles were performed.Obtain specific target genes for differential miRNAs.GO function and KEGG signal pathway enrichment analysis were performed on target genes of differential miRNAs using Gene Ontology and KEGG databases.To construct miRNA-m RNA regulatory network for acupuncture against asthma,and to locate key miRNAs that play a central role in acupuncture Antiasthmatic.Based on the key miRNAs,constructed the miRNA-Gene-GO regulatory network,and the key regulatory target genes of acupuncture anti-asthma effect were screened out,and analyze its role in the anti-asthma process of acupuncture and acupuncture in "Three-acupoint and Five-needle Method ".Results:1.According to the difference expression significance standard,it was determined that there were 22 significant differentially expressed miRNAs and 1381 significantly differentially expressed m RNAs in the asthma patients before and after treatment with “Three-acupoint and Five-needle Method” acupuncture.2.miRNA target gene prediction results showed that 3101 target genes were up-regulated by miRNA,and 3869 target genes were down-regulated.Bioinformatics analysis based on predicted target genes showed that up-regulated target genes mainly involved in metabolism and stimulation,and humoral immunity.Biological processes,involved in significant signaling pathways including metabolic pathways,MAPK,P53,Fox O and other signaling pathways;down-regulated target genes are mainly involved in immune processes,regulation of metabolic processes and stimuli,and other biological processes,including significant signaling pathways including Wnt,PI3K-Akt,T cell receptor,TNF-?and other immune and inflammation signaling pathways,the biological processes involved in these genes and signal transduction pathways play an important role in asthma diseases.3.The combined analysis of miRNA microarray and m RNA microarray showed that up to 136 specific target genes were up-regulated by miRNA regulation after needle acupuncture.The salient functions of these specific target genes are mainly zinc ion transmembrane transport,steroid hormone response,phosphoprotein phosphatase activity regulation,calcium ion signaling regulation and the like.Significant signaling pathways involved in natural killer cell-mediated cytotoxicity,inflammatory mediator regulation of TRP channels,phosphatidylinositol signaling system,PI3K-Akt signaling pathway,p53 signaling pathway,inositol phosphate metabolism,etc.4.Through the construction of miRNA-m RNA regulatory network,it was found that mi R-6085,mi R-374a-5p,mi R-7975,mi R-30e-3p,mi R-6749-5p,mi R-30c-5p play a central role in acupuncture anti-asthma effect Key miRNAs for regulation.5.Based on the miRNA-Gene-GO regulatory network constructed by acupuncture anti-asthmatic key miRNAs,WNT10 B,UBE3A,NR2F2,CCND1,PGR,CALM1,VDR,HTT,ANXA1 and CLU are the main target genes for acupuncture.Epithelial cell proliferation regulation,zinc ion transmembrane transport,ATPase activity regulation,T cell differentiation regulation,protein ubiquitination regulation,second messenger-mediated signaling,calcium signaling,regulation of phosphoprotein phosphatase activity,The reaction of steroid hormones and the like is a biological process in which acupuncture is mainly regulated by the anti-asthmatic effect.Conclusion:1.miRNA microarray data analysis Screened that 22 differentially expressed miRNAs related to acupuncture anti-asthma effect,combined with m RNA chip analysis to identify 192 specific target genes.2.Bioinformatics analysis of miRNA and its specific target genes revealed that mi R-6085,mi R-374a-5p,mi R-7975,mi R-30e-3p,mi R-6749-5p,mi R-30c-5p,etc.miRNAs that play a central role in the anti-asthma effect.3.Through the construction of miRNA-Gene-GO regulatory network,it was found that WNT10 B,UBE3A,NR2F2,CCND1,PGR,CALM1,VDR,HTT,ANXA1 and CLU are the main target genes for acupuncture regulation.4.Acupuncture may be regulated by epithelial cell proliferation,zinc ion transmembrane transport,ATPase activity regulation,T cell differentiation regulation,protein ubiquitination regulation,second messenger-mediated signaling,calcium signaling,phosphoprotein Biological regulation mechanisms such as regulation of phosphatase activity and response to steroid hormones exert anti-asthmatic effects.
